作者盛守新(2019)在《乳酸菌胞外多糖佐剂提高金黄色葡萄球菌疫苗免疫效果的研究》一文中研究指出:金黄色葡萄球菌(Staphylococcus aureus,S.aureus)是一种人畜共患病原菌,可引起人和动物的多种感染。同时,S.aureus也是最常见的细菌性食物中毒病原之一。近些年,耐甲氧西林金黄色葡萄球菌广泛流行,目前S.aureus的感染面临“无药可治”的严峻挑战。因此,加强对S.aureus感染的免疫防治研究,研制高效、安全的新型金葡菌疫苗具有重要的经济意义和公共卫生学意义。多年来,全世界科研工作者对金葡菌疫苗研制进行了积极的探索,然而,目前尚无有效疫苗问世。许多学者认为,目前金葡菌疫苗的研究忽略了S.aureus对天然免疫系统的削弱和破坏作用,并进一步影响后续获得性免疫系统功能的发挥。采用有效的疫苗佐剂配伍,激发更强的免疫应答应该是金葡菌疫苗研制过程中要考虑的问题。然而,现有的免疫佐剂引起的免疫应答效应相对单一,极大的限制了疫苗免疫保护作用的充分发挥,因此研发有效、安全的新型免疫佐剂是未来金葡菌亚单位疫苗研发的重要发展方向。乳酸菌胞外多糖(Lactic acid bacteria exopolysaccharide,LAB EPS)是从乳酸菌发酵液中分离出来的活性成分。实验室前期研究证明EPS可以增强巨噬细胞免疫活性,促进树突状细胞成熟和前炎性因子分泌,并且可以显著提高小鼠对OVA抗原Th1型和Th17型免疫应答。本研究在实验室前期研究基础上,选取两种实验室分离的乳酸菌胞外多糖(EPS30和EPS29)配伍金黄色葡萄球菌3S疫苗(混合FhuD2、HlaH35L和EsxA-EsxB抗原)免疫小鼠,观察其在小鼠感染金黄色葡萄球菌菌血症模型和乳房炎感染模型中的免疫保护作用,为金黄色葡萄球菌疫苗佐剂的研究与开发提供新思路。本研究主要内容如下:1.分别制备金黄色葡萄球菌3S疫苗和乳酸菌EPS30及EPS29多糖,将EPS30和EPS29佐剂分别与3S疫苗混合后免疫小鼠。检测免疫小鼠抗原特异性IgG抗体水平、T淋巴细胞增殖能力以及脾脏T淋巴细胞的细胞因子表达情况。结果发现,EPS29和EPS30可以显著提高抗原特异性IgG抗体水平和T淋巴细胞增殖能力。同时,EPS29可以显著提高小鼠脾脏中CD4+IFN-γ+T细胞、CD4+IL-4+T细胞和CD4+IL-17+T细胞比例。2.通过尾静脉攻毒金黄色葡萄球菌,建立小鼠菌血症感染模型。观察各免疫组在感染金黄色葡萄球菌后的存活率、体重变化、各脏器细菌定植量及病理变化,对乳酸菌胞外多糖佐剂配伍3S疫苗在小鼠菌血症感染金葡菌模型的保护效果进行评价。结果发现,相比单独免疫金葡菌3S抗原,EPS29佐剂能有效提高小鼠感染金黄色葡萄球菌后的生存率,并能显著减少金葡菌在小鼠各脏器的定植,减轻小鼠各脏器病变。3.通过乳腺感染金黄色葡萄球菌,建立小鼠乳腺炎感染模型。检测免疫小鼠抗原特异性IgG抗体水平、观察小鼠乳腺炎临床症状、细菌在乳腺组织定植量、病理变化及炎性因子IL-6、TNF-α和IL-1β的表达水平,对乳酸菌胞外多糖佐剂配伍3S疫苗在小鼠乳腺感染金葡菌模型的保护效果进行评价。研究发现EPS29作为免疫佐剂,与3S抗原混合免疫小鼠,可以显著提高抗原特异性IgG抗体水平。与对照组相比,EPS29佐剂能显著减少小鼠乳腺内细菌的定植和减轻小鼠乳腺病变,降低乳腺中IL-6和TNF-α的含量。上述结果提示:乳酸菌胞外多糖佐剂EPS29配伍金黄色葡萄球菌3S疫苗可以有效的保护小鼠金黄色葡萄球菌全身感染和乳房感染,为新型有效的金黄色葡萄球菌疫苗佐剂的设计提供了实验基础与理论指导。
Abstract
