作者刘春晓(2019)在《拟南芥中AtR8 lncRNA功能及作用机制初探》一文中研究指出:非编码RNA(non-coding RNA)不翻译成蛋白质,它们通过转录、转录后及翻译水平调控靶基因表达,在生物体生长发育过程中发挥重要作用。大于200个核苷酸长度的为长非编码RNA(long non-coding RNA,IncRNA),它们以调节因子形式,在植物开花、雄性不育、营养代谢、生物和非生物胁迫等过程中起着重要作用。AtR8lncRNA(259 nt)是 2012 年于拟南芥(Arabidopsisthaliana)中发现的一个 RNA 聚合酶IⅢ转录的lncRNA,种子萌发期其于拟南芥根尖端大量表达,并响应ABA、NaCl、GA3、SA、Mannitol等应激环境;其同源物存在于结球甘蓝(Brassica oleracea L.var capitat L.)和白菜(Brassica rapa ar pekinensis)等十字花科芸薹属植物中。目前,AtR8 lncRNA的详细功能及作用机制尚不清楚。本研究表明,正常条件下,atr8 突变体导致种子干瘪、长角果发育不完全、莲座叶边缘锯齿状、成熟植株纤细矮小、种子萌发率大幅度降低,这些表型在AtR8 lncRNA过表达(AtR8-AtOX)植株中没有被观察到。ABA应激条件下,atr8突变体和AtR8-AtOX均抑制了种子萌发。分子水平分析表明,正常萌发和ABA应激处理下,atr8突变体和AtR8-AtOX在萌发期间均影响了 ABA信号径路中萌发相关重要基因表达。本研究也深入分析了结球甘蓝中AtR8 lncRNA的同源物BoNR8 lncRNA(272nt)的功能及作用机制。我们研究结果表明,BoNR8 lncRNA主要于结球甘蓝萌发种子根伸长区表皮层大量表达,响应ABA和NaCl应激。正常条件下,拟南芥中BoNR8 lncRNA过表达主要通过影响AtRAV1基因表达抑制了种子萌发、角果形成、根生长。ABA应激下,BoNR8 lncRNA主要通过影响ABA信号径路中WRKY6、AARV1、RD29、SnRK2.3等萌发相关重要基因表达降低了萌发种子对ABA的敏感。
Abstract
