作者李飞(2019)在《阴道毛滴虫诱导THP-1细胞产生胞外陷阱的研究》一文中研究指出:阴道毛滴虫在世界范围内分布广泛感染率较高,可能是最常见的通过性传播的人兽共患原虫。阴道毛滴虫一般寄生在人体内,也可以感染猴等动物。人感染阴道毛滴虫后,会产生阴道炎及尿道炎等症状,并且阴道毛滴虫感染与艾滋病病毒、乳头瘤病毒、单纯疱疹病毒的感染有关[1]。胞外陷阱对机体抵御病原入侵具有重要作用,已有研究发现牛艾美尔球虫、新孢子虫、弓形虫等原虫刺激免疫细胞产生了胞外陷阱。胞外陷阱可以捕获入侵的原虫,并且有些胞外陷阱对原虫具有杀伤作用[2]。贾第虫也可刺激免疫细胞产生胞外陷阱[3],但是同为鞭毛虫的阴道毛滴虫是否能刺激细胞产生胞外陷阱并释放活性氧(ROS)等物质阻碍病原入侵,目前尚未见报道。THP-1细胞是人外周血单核细胞系,本试验应用扫描电子显微镜观察了阴道毛滴虫刺激THP-1细胞产生的胞外陷阱;应用激光共聚焦检测了阴道毛滴虫刺激THP-1细胞产生的胞外陷阱的成分;应用活性氧检测试剂盒检测了阴道毛滴虫刺激THP-1细胞产生胞外陷阱时ROS的水平;应用乳酸脱氢酶(LDH)检测试剂盒检测了阴道毛滴虫刺激THP-1细胞产生胞外陷阱时LDH的水平;应用Picogreen双链DNA检测试剂盒检测了刺激时间、刺激比例对阴道毛滴虫刺激THP-1细胞产生胞外陷阱的影响;应用Picogreen双链DNA检测试剂盒检测了不同状态的阴道毛滴虫对阴道毛滴虫刺激THP-1细胞产生胞外陷阱的影响;应用Western blot检测了阴道毛滴虫刺激THP-1细胞产生胞外陷阱时P38MAPK、ERK1/2MAPK信号通路的磷酸化水平;应用IL-1β、TNFα细胞因子检测试剂盒检测了阴道毛滴虫刺激THP-1细胞产生IL-1β、TNFα的水平。扫描电子显微镜结果表明阴道毛滴虫刺激THP-1细胞产生了胞外陷阱,阴道毛滴虫刺激THP-1细胞产生了细长的、具有多个网孔的纤维样网状结构缠绕并捕获了阴道毛滴虫,这直接有效的证明了阴道毛滴虫刺激THP-1细胞产生了胞外陷阱;激光共聚焦结果表明阴道毛滴虫刺激THP-1细胞产生的胞外陷阱具有胞外陷阱的典型结构特征:DNA骨架上镶嵌着组蛋白(H3)、髓过氧化物酶(MPO);ROS检测结果表明胞外陷阱产生时产生了活性氧,这证明胞外陷阱具有杀伤病原的功能;LDH检测结果表明胞外陷阱产生时THP-1细胞并未发生裂解,这证明胞外陷阱是一种不同于细胞坏死的新的细胞死亡方式;DNA检测结果表明阴道毛滴虫刺激THP-1细胞产生的胞外陷阱具有剂量依赖性,随刺激时间延长逐渐减少,胞外陷阱的产生与阴道毛滴虫的活力没有关系;Western blot检测结果表明阴道毛滴虫刺激THP-1细胞产生胞外陷阱时P38MAPK、ERK1/2MAPK信号通路磷酸化水平上升,这证明了阴道毛滴虫刺激THP-1细胞产生的胞外陷阱与P38MAPK、ERK1/2MAPK信号通路有关;IL-1β、TNFα检测结果表明阴道毛滴虫刺激THP-1细胞产生了IL-1β、TNFα,这证明阴道毛滴虫刺激THP-1细胞产生的胞外陷阱可能与IL-1β、TNFα有关,本研究为探究阴道毛滴虫刺激THP-1细胞产生胞外陷阱的机理提供了新思路。
Abstract
