作者陈强(2019)在《猕猴桃AcPDS1基因对番茄果实品质的影响》一文中研究指出:番茄果实的成熟发育伴随着众多代谢物质的合成与分解,在这些代谢途径中,类胡萝卜素在番茄体内的合成是最为重要的过程之一。类胡萝卜素是在自然界中广泛存在的一种有机色素,类胡萝卜素具有多种对人体有益的功能。由于其在体外易分解的特性,我们只能通过进食果蔬来摄取人体所必需类胡萝卜素。番茄果实类胡萝卜素的生物合成过程中涉及众多基因调控,每个参与调控的基因都起着各自不可或缺的作用。这其中PDS(八氢番茄红素脱氢酶)调控八氢番茄红素脱氢形成番茄红素,是合成过程中重要的限速酶。本文克隆了猕猴桃AcPDS1基因,进行了如下研究工作:(1)构建了植物瞬时表达载体,利用农杆菌注射的方法在烟草叶片中瞬时表达,并通过Western blot技术,检测在烟草叶片中猕猴桃PDS基因的表达情况。(2)构建了AcPDS1基因过表达载体,通过农杆菌介导稳定遗传转化番茄,获得转基因番茄植株。通过表型分析,代谢产物测定,类胡萝卜素生物合成相关基因的表达情况,来阐述AcPDS1转基因番茄植株的相关表型。试验数据表明:(1)利用构建的AcPDS1基因植物瞬时表达载体侵染烟草,在烟草叶片中检测到与预期大小相同的PDS蛋白,且含量大大增加;(2)利用液相色谱技术检测到瞬时表达的烟草叶片β-胡萝卜素含量明显上升;(3)通过激光共聚焦定位AcPDS1蛋白的表达位置在细胞膜和细胞质中;(4)构建的AcPDS1基因过表达载体,通过农杆菌侵染野生型番茄叶片和茎,筛选分化,得到了AcPDS1转基因番茄植株;(5)通过液相色谱技术检测,相较于野生型番茄果实,AcPDS1转基因植株果实内的番茄红素及β-胡萝卜素含量均有提高;(6)AcPDS1转基因植株中,在类胡萝卜素合成过程中起作用的合成相关基因、乙烯信号应答基因表达量也受到影响。本工作为进一步探讨AcPDS1基因在类胡萝卜素合成中的所起的具体分子机制提供了良好的基础。
