作者张佳斌(2019)在《江蓠琼脂提取与顺丁烯二酸酐酯化琼脂制备工艺及其应用》一文中研究指出:琼脂是一种天然水溶性高分子多糖,因其具有独特的凝固性和凝固稳定性,常被用做增稠剂、乳化剂、悬浮剂和凝胶剂,广泛应用于食品、日化、医药化工等领域。为解决琼脂在碱法提取过程中胶质流失、得率低、污染大以及琼脂凝胶透明度低而造成其应用范围窄等问题,本文在碱法、酶法和酶法辅助提取工艺基础上,分析三种提取工艺过程中江蓠藻体变化趋势,为指导琼脂生产提供理论依据;计算江蓠藻损失率、琼脂得率、考察琼脂指标差异,为提高琼脂得率、减少生产损耗提供指导。然后以顺丁烯二酸酐为酯化剂对琼脂进行湿法改性,旨在保证琼脂基本性质的前提下提升琼脂凝胶透明度,为琼脂衍生物制备方法提供思路。最后将改性琼脂应用到微胶囊技术(作为壁材)领域,拓宽琼脂及其衍生物应用范围。首先,以未处理江蓠藻做参照,对碱法、酶法和酶法辅助提取工艺进行比较,考察三种提取工艺过程对江蓠藻体、江蓠损失率、琼脂得率等各项指标的差异。研究发现:纤维素酶只酶解、软化细胞壁,不会作用于其他物质;碱处理会破坏细胞壁,溶解胶质物,可作用于多糖分子脱除硫酸基团形成3,6-内醚-半乳糖,提高琼脂凝胶强度;酸、漂白处理会进一步软化江蓠,便于琼脂的提取。随着琼脂提取工艺过程的进行,江蓠藻体逐渐被破坏,原有的厚质细胞壁被去除,裸露出包裹胶质的皮层细胞;碱法提取工艺中,江蓠藻损失率为63.0%、琼脂得率为12.3%,凝胶强度为760.6 g/cm~2。与碱法提取工艺相比,酶法提取工艺(酶处理、酸处理和漂白处理)未经碱处理,硫酸基团含量较高(3.4%),凝胶强度低(358.1 g/cm~2)、琼脂得率高(28.5%)、江蓠藻损失率低(22.6%)。与碱法提取工艺相比,酶法辅助工艺(酶处理、碱处理、酸处理和漂白处理)采用纤维素酶对江蓠藻进行预处理,江蓠藻损失率降低(45.9%),琼脂得率升高(20.3%),低浓度碱(3%)处理后,硫酸基团下降(1.9%),凝胶强度增加(695.7 g/cm~2)。碱法提取工艺所得琼脂的白度、透明度、溶点、凝固温度和融化温度高于酶法辅助和酶法提取工艺;三种提取工艺所得琼脂粘度(18~20 cP)均高于未处理样品(9.3 cP)。其次,以顺丁烯二酸酐为酯化剂,氢氧化钠为催化剂,对琼脂进行湿法改性,以获得高透明度的顺丁烯二酸酐酯化琼脂。通过单因素实验得到最佳制备工艺为:顺丁烯二酸酐添加量2%、反应pH 7-7.5、反应温度30℃、琼脂浓度9%、滴加时间1 h、继续反应时间0 h。最佳工艺条件下,顺丁烯二酸酐酯化琼脂透明度从51%提升至79%,凝胶强度为867.4g/cm~2;溶胀性、乳化性改善;白度、3,6-内醚-半乳糖、硫酸基团、凝固温度、融化温度和溶解温度略有下降。FTIR测试结果显示顺丁烯二酸酐酯化琼脂出现新的酯基吸收峰,顺丁烯二酸酐与琼脂酯化成功;TG测试结果显示顺丁烯二酸酐酯化琼脂稳定性下降,保水性增加。最后,采用锐孔-凝固浴法,以顺丁烯二酸酐酯化琼脂为微胶囊壁材,大豆油为包埋芯材,制备酯化琼脂微胶囊。通过单因素实验得到最佳的酯化琼脂微胶囊制备工艺为:酯化琼脂(壁材)浓度1.5%、芯材含量8 mL、干燥温度60℃、鼓风干燥方式。最佳工艺条件下,酯化琼脂微胶囊粒径为0.14 cm、载油率为56.3%、包埋率99.1%。在此工艺条件下,对DHA进行微胶囊化,对比不同芯材的乳液稳定性,得到粒径为0.15 cm、载油率为62.2%和包埋率为99.2%的酯化琼脂-DHA微胶囊。加速氧化7 d试验结果显示,酯化琼脂-DHA微胶囊过氧化值(67.4 mmoL/kg)明显低于未包埋DHA(860.1 mmoL/kg)的过氧化值,酯化琼脂-DHA微胶囊贮藏稳定性显著提高。
