作者涂小艳(2019)在《刺梨渣中种子油提取及其在囊泡制备上的应用研究》一文中研究指出:贵州刺梨酒生产中会产生大量的刺梨果渣,从果渣种子中提取的种子油富含亚油酸、亚麻酸等不饱和脂肪酸;不饱和脂肪酸囊泡在药物包埋和缓释方面都有重要应用。从将刺梨果渣变废为宝、节约资源的角度考虑,研究以贵州刺梨酒生产中的果渣为原料,进行种子油提取,利用尿素包合法纯化种子油中亚油酸及亚麻酸,将纯化后的脂肪酸用于混合脂肪酸囊泡的制备,最后应用于抗癌药物阿糖胞苷的缓释。研究可为刺梨果渣的回收利用提供新的思路。本文以刺梨果渣中种子为原料,采用微波法对刺梨种子油进行提取,通过响应面法,以出油率作为评价指标,对提取工艺进行优化;通过尿素包合法纯化种子油中的亚油酸及亚麻酸,利用气相色谱对种子油中的亚油酸、亚麻酸含量进行测定。根据回归分析,确定各因素对提取工艺的影响次序依次为:超声波>提取时间>液料比;求得的最佳提取参数为:液料比1:11,微波功率400w,提取时间40min。经验证实验平均出油率为12.51%,理论预测值与试验结果接近,说明优化工艺参数可信度高,优化工艺参数可信度高,具有参考价值。气相结果表明,未进行尿素包合前,刺梨种子油中亚油酸和亚麻酸总含量为26.52%(其中亚油酸含量为17.93%,亚麻酸含量为8.59%);进行尿素包合纯化后,亚油酸和亚麻酸的含量都提高了三倍以上,两者总含量为97.7%(亚油酸含量为65.38%,亚麻酸含量为32.32%),达到了纯化的目的,纯化前后混合脂肪酸(亚油酸及亚麻酸)比例保持不变,大约为2:1。将纯化后的混合脂肪酸作为脂肪酸囊泡的模板脂肪酸,采用表面张力法研究其在不同pH条件下的自组装行为,并采用酸碱平衡滴定实验分析混合脂肪酸自组装体形成囊泡的pH范围,采用动态光散色(DLS)分析其粒径分布,透射电子显微镜(TEM)观察其形貌。实验结果表明,当pH在13.0~9.0时,体系溶液外观清澈透明,混合脂肪酸主要以离子状态存在,离子间的静电排斥力使其自组装成胶束;当pH在9.0~8.0时,体系溶液外观表现为强烈乳光现象,混合脂肪酸分子与离子间的数量相当,两者以氢键作用力使其自组装成囊泡;当pH<8.0后,体系溶液外观浑浊甚至油水分离,溶液中主要是混合脂肪酸分子,使囊泡自组装体解体。用透射电镜和动态光结果表明pH 10.0时获得9 nm量级的球形胶束结构,pH 9.0时获得22 nm左右的囊泡结构,pH 8.0时获得40nm左右的类囊泡结构,说明混合脂肪酸能够发生pH响应自组装形成胶束或者囊泡等各种形貌和尺度的自组装体结构。混合脂肪酸囊泡对pH依赖性强,pH窗口很窄并且偏离生命体系适应的pH范围,限制其在药物包埋/缓释中的应用。为了迁移与扩张混合脂肪酸囊泡pH窗口,选择表面活性剂吐温80(Tween80)作为模板表面活性剂,加入到混合脂肪酸体系中,将pH窗口迁移至生命体系所适应的范围内。实验结果表明,吐温80加入到混合脂肪酸体系中,将囊泡的窗口由原来的pH 8.0~9.0迁移到pH 5.5~7.5,囊泡的pH窗口落在了生命体系所适应的pH范围内。由于吐温80中含有聚氧乙烯基(PEO)结构,能够在酸性条件下与混合脂肪酸中脂肪酸分子间产生氢键作用力,代替了原先在碱性范围内脂肪酸分子与离子间的氢键作用力,同时由于吐温80有增溶效果,增大混合脂肪酸在酸性条件下的溶解性,使混合脂肪酸囊泡在更宽的pH范围和更低的pH环境下形成并稳定。囊泡的粒径会随着pH的降低而增大,这是由于pH的变化将导致体系中形成囊泡的自组装单元发生变化,pH越低,混合脂肪酸体系中羧酸离子越少,羧酸离子间的静电排斥力减小,形成的囊泡结构越紧密,囊泡粒径就越大。将加入了吐温80的混合脂肪酸囊泡包覆抗癌药物阿糖胞苷,研究其包封率及其在人工肠液中的释放情况。研究结果表明,混合脂肪酸/吐温80复合囊泡对阿糖胞苷的包封率为42.07%;在人工肠液中,时间为9h,囊泡包封的阿糖胞苷释放率是40%,游离阿糖胞苷的释放率是80%,说明混合脂肪酸/吐温80复合囊泡对阿糖胞苷具有明显的缓释作用。将混合脂肪酸/吐温80复合囊泡与亚油酸/吐温80复合囊泡、亚麻酸/吐温80复合囊泡进行对比,发现三种复合囊泡的包封率及缓释效果相当。亚油酸和亚麻酸的分离过程比较复杂,费钱费力,如果不将二者分离,直接应用,可以简化工艺,提高经济效应。刺梨种子油中的亚油酸和亚麻酸纯化工艺简单,将纯化后的混合脂肪酸直接制备囊泡,应用于药物包埋/缓释,操作简单可行,具有很好的理论和实际应用价值。
