作者王琛(2019)在《黄瓜根发育异常突变体及其候选基因的鉴定》一文中研究指出:黄瓜(Cucumis sativus L.2n=2x=14)为葫芦科甜瓜属植物,是世界上主要的蔬菜作物,也是我国广泛栽培的设施蔬菜作物之一。根作为植物的营养器官,在植株整个生长发育历程中发挥着至关重要的作用。根是黄瓜植株的重要组成部分,具有吸收营养物质和水分等功能。黄瓜根发育异常表型是研究植株根发育机理及培育优良品种的理想性状,但是相关研究报道较少。本研究在前期构建黄瓜EMS诱变突变体库基础上构建了黄瓜EMS诱变根发育异常突变体‘Csrod(Cucumis sativus root dysplasia)’的M3代遗传群体,对其进行表型鉴定,并利用MutMap+技术对突变表型和野生表型混池分别进行重测序分析,对根发育异常候选基因进行了初步定位,运用KASP基因分型技术对候选SNP在M3代群体中进行了鉴定,并最终确定了黄瓜EMS诱变根发育异常候选基因。主要结果如下:(1)表型鉴定结果表明,黄瓜根发育异常突变体‘Csrod’在根发育等方面与野生型存在显著差异。根发育异常突变体‘Csrod’根部较野生型生长慢、只能生长至1-2 cm,植株生长至14 d左右,根与子叶分离,为致死表型。(2)为了鉴定黄瓜根发育异常突变体‘Csrod’候选基因,将M2代野生型表型单株进行自交得到M3代群体,其中一个有性状分离的M3群体中有11株为突变体表型单株,93株表现为野生型表型。利用MutMap+方法进行极端性状混池重测序分析,选取M3代中突变表型单株11株和野生型表型单株34株,提取等量DNA进行混合,分别构建突变体表型池和野生型表型池进行混池重测序,并与黄瓜基因组序列进行比对,依据SNP-index以及突变位点对基因的影响进行综合分析,筛选到位于黄瓜第3号染色体的5个候选SNP,其中4个候选SNP位于基因外显子区,另外1个候选SNP则位于终止密码子区。结合KASP基因分型技术,在M3代群体中发现基因ID为Csa3G104930包含的SNP的基因型与M3群体单株表型共分离,表明Csa3G104930为调控黄瓜根发育异常的候选基因。序列分析结果表明Csa3G104930基因全长3876 bp,编码831个氨基酸,参与3-脱氧-D-甘露糖-八磺酸盐合成酶及酰基神经氨酸蛋白家族的胞苷酰基转移酶的合成,因此将黄瓜根发育异常突变体候选基因命名为Cscks(Cucumis sativus CMP-KDO synthetase)。本研究初步鉴定出一个参与调控黄瓜根部发育的新基因,并对其基因功能进行了初步探讨,下一步将对该基因功能及分子机制进行深入研究,进而对黄瓜营养生长、新品种培育等研究提供参考。
Abstract
