作者吴会杰(2019)在《SLCCNV和MNSV侵染性克隆的构建及致病因子鉴定》一文中研究指出:甜瓜(Cucumis melo)是我国重要的经济作物之一。近年来,新突发病毒病严重影响甜瓜产业发展。中国南瓜曲叶病毒(squash leaf curl China virus)和甜瓜坏死斑点病毒(melon necrotic spot virus)是中国新近报道为害甜瓜的种。为阐明两种病毒的致病机理,构建了SLCCNV和MNSV两种甜瓜病毒的侵染性克隆,分析了病毒编码的病毒致病因子,以期为防控该病毒病奠定基础。目前取得以下结果:1、鉴定了甜瓜是SLCCNV的新寄主,构建了SLCCNV侵染性克隆并进行了烟粉虱传染试验。测定了SLCCNV-HN全基因组序列并进行了系统进化分析,结果表明SLCCNV-HN甜瓜分离物与SLCCNV-Hn南瓜分离物、SLCCNV-Hn61和SLCCNV-SY西葫芦分离物聚为一类。构建了SLCCNV-HN侵染性克隆载体,接种甜瓜后植株矮化、叶片皱缩。进行了烟粉虱传染试验,发现烟粉虱接种的植株表现出与接种野生型一致的症状。2、明确了AC5基因是SLCCNV的致病因子,证实了AC5是SLCCNV编码的沉默抑制子,其沉默抑制活性与核定位信号相关。将表达AC5基因的PVX异源载体接种本氏烟后叶片表现枯斑、皱缩,表明AC5是致病因子并激发了植物的过敏性坏死反应,DAB染色说明过敏性坏死反应与H2O2的积累相关。不仅如此,利用SLCCNV侵染性克隆定点突变AC5基因,进一步证实了AC5基因是该病毒的致病因子。利用p35S-AC5与p35S-GFP共浸润接种16c本氏烟,接种5 d后接种部位出现明显的绿色荧光信号,表明了AC5基因可抑制局部沉默。利用p35S-GFP、p35S-dsGFP和p35S-AC5共浸润野生本氏烟,接种5 d后在接种部位未发现绿色荧光信号,表明AC5不抑制双链GFP(IR-PTGS)诱导的沉默。在AC5基因ORF之前引入终止密码子,明确了AC5是在蛋白水平发挥作用。利用AC5核定位信号缺失突变体与p35S-GFP共浸润16c本氏烟,发现该突变体失去了沉默抑制作用,表明AC5蛋白的核定位信号与沉默抑制活性相关。3、证实了p42是MNSV的致病因子,与寄主因子3-羟基异丁酸脱氢酶(3-Hydroxyisobutyrate dehydrogenase-like 1,3HID1)互作。将表达p42基因的PVX异源载体接种本氏烟后植株新叶枯死,证实p42是MNSV的致病因子。构建了MNSV侵染性克隆,在此基础上构建了携带GFP的MNSV侵染性克隆载体。在MNSV侵染性克隆的基础上进行了p42基因突变,发现p42的R-domain和S-domain是致病功能域。此外,构建了受MNSV侵染的甜瓜cDNA文库,筛选到与p42互作的6个寄主因子,经酵母双杂交、BIFC及共定位一对一验证,表明3HID1与p42互作,本研究为揭示p42与寄主互作的机制奠定了基础。
