作者刘玲(2019)在《石墨烯量子点-稀土氟化物复合材料的制备及光学性能研究》一文中研究指出:稀土元素可以通过4f或4f-5d跃迁表现出强烈的荧光发射,与普通元素相比具有无可比拟的光谱特性,稀土发光材料也因其极具特色的发光性质受到了热切关注。其中稀土上转换发光材料采用近红外光激发,具有辐射损伤低、穿透能力强、自体荧光弱等优势,被认为是新一代的生物光学探针。目前所合成的稀土发光材料存在功能性不强、上转换发光效率不高等问题,限制了它在生物医学领域的应用。石墨烯量子点是另一种极具前景的纳米光学材料,具有水溶性好、易功能化的特点。然而,石墨烯量子点通常采用紫外光激发产生可见光,会产生大的背景荧光干扰且对生物体损伤大,导致应用受限。因此,将石墨烯量子点与稀土发光材料有效复合,实现它们在性能上的“优势互补”,并对两者之间的作用机理开展研究具有重要的现实意义。以柠檬酸为碳源、组氨酸为功能化试剂,通过一步热解法制备组氨酸功能化石墨烯量子点。合成的石墨烯量子点片边缘含有丰富的咪唑基团,可以与稀土金属离子形成稳定配合物,为之后复合物的制备奠定了基础。继续引入己二胺功能化试剂,制备了组氨酸-己二胺双功能化石墨烯量子点。研究表明,制备的石墨烯量子点尺寸约2.9 nm,厚度为1-2层石墨烯层。己二胺的引入使得石墨烯片边缘含有大量氨基。氨基具有较强的供电子效应,可提高石墨烯片中的电荷密度,有利于提高石墨烯量子点的发光效率。此外,氨基可通过酰胺化反应与羧基化DNA偶联,为进一步在生物领域应用提供了便利。以组氨酸功能化石墨烯量子点为稳定剂水热合成了石墨烯量子点@NaTbF4复合材料。研究表明,NaTbF4为六方相晶体,石墨烯量子点以共价键方式包覆于NaTbF4表面,粒子尺寸仅为4-6 nm。小的尺寸和丰富的亲水基团使它易分散于水中,分散液具有优异的稳定性。NaTbF4吸收光谱与石墨烯量子点发射光谱存在较大重叠,且距离极短,它们的复合诱导了显著的荧光共振能量转移。与单独的石墨烯量子点相比,石墨烯量子点@NaTbF4复合材料的荧光强度增加超过7倍。复合物材料能与Cu2+配位产生明显的荧光猝灭,猝灭的荧光又可被组氨酸恢复。基于以上行为,建立一种“开-关”型组氨酸荧光检测方法。组氨酸浓度在1.0×10-6-2.0×10-4mol·L-1之间峰荧光强度随组氨酸浓度的增加而线性增大。方法检出限达到3.8×10-7mol·L-1(S/N=3),对无机离子和天然氨基酸有良好的选择性,成功用于人体尿液中组氨酸检测和宫颈癌细胞(Hela细胞)成像。以组氨酸-己二胺双功能化石墨烯量子点为稳定剂水热合成了石墨烯量子点@NaYF4:Yb,Er纳米复合物。结果表明,复合物具有良好的水溶性,粒径为11.5 nm。石墨烯量子点对近红外激发光有较强吸收能力,可作为“天线”集中捕获光子能量并转移给上转换纳米粒子,导致上转换发射显著增强。将复合物光学探针用于构建检测癌胚抗原的上转换生物传感纳米平台。首先,连接DNA(LDNA)共价连接到复合物表面。其次,发夹DNA(Hp)修饰的金纳米粒子与LDNA修饰的复合物通过DNA杂交化反应形成缀合物,二者之间发生荧光共振能量转移,有效猝灭了复合物的上转换发射。最后,纳米缀合物上的Hp被信号DNA(SDNA)特异性打开,导致猝灭的上转换荧光发射迅速恢复。此外,利用核酸外切酶Ⅲ辅助的目标循环放大策略生成SDNA,同时释放癌胚抗原。被释放的癌胚抗原能够继续参与循环产生更多SDNA,使得信号显著放大。该生物传感器在检测癌胚抗原方面具有良好的分析性能,线性范围为5.0×10-17-1.0×10-8g·mL-1,检测限低至2.4×10-17g·mL-1(S/N=3),且具有较高的特异性,成功用于人血清中癌胚抗原的分析检测。
