作者吕阳欧(2019)在《犬小孢子菌转录组测序分析及Zrta基因转化株的构建》一文中研究指出:犬小孢子菌(Microsporum Canis)是一种常见的人畜共患皮肤癣菌,主要感染皮肤、毛发和甲等角化组织,引起浅部真菌病。在真菌的生长和繁殖中,锌是一种必须微量元素,参与许多生物过程,对真菌的生长和代谢具有重要作用。在病原真菌与宿主相互作用过程中,宿主的营养免疫限制病原真菌对锌的摄取,为了能在锌限制条件下存活和增殖,病原真菌进化出特殊的锌稳态调控机制。锌响应激活蛋白(Zap1)及其同系物是调节锌稳态基因的主要转录因子,在酿酒酵母、烟曲霉、念珠菌等真菌中已被证实。但目前锌缺乏对犬小孢子菌基因表达的影响尚不清楚,关于犬小孢子菌锌响应激活蛋白参与的生物过程尚不明确。确定锌缺乏对犬小孢子的影响,明确锌响应转录激活因子在犬小孢子菌锌稳态调节中的作用,对犬小孢子菌锌稳态调控的相关研究具有重要意义。本试验通过对正常组(NORM)和锌限制条件下(Zn200和Zn800)生长的犬小孢子菌进行转录组测序,对所得数据进行的原始数据质控、比对分析、表达定量分析、差异表达分析、差异表达基因的功能富集和注释等,明确锌缺乏对犬小孢子菌基因表达的影响。通过根癌农杆菌介导的遗传转化(Agrobacterium tumefaciens-mediated transformation,ATMT)构建了犬小孢子菌锌响应转录激活因子Zrta基因缺失株,为后续Zrta基因功能分析奠定基础。研究结果如下;1.获得大量基因数据,差异表达基因数目统计显示,Zn800组和Zn200组与NORM组相比,上调基因分别为798、764个,下调基因分别为585、631个,其中MCYG04486、MCYG06235在锌限制组与正常组相比中均显著上调,MCYG02504只在Zn200中显著上调;Zn800组与Zn200组相比上下调基因的数目分别为3个和8个。差异表达基因功能注释分析表明MCYG06235(zinc-responsiveness transcriptional activator)可能为重要的锌转运因子。2.高保真扩增了犬小孢子菌锌响应转录激活因子基因MCYG06235(zinc-responsiveness transcriptional activator,Zrta),构建了含有Zrta基因两侧同源片段及潮霉素B抗性基因(hph)的双元载体pDHt/ZrtaⅠ、Ⅱ::hph,酶切和PCR验证证实双元载体构建成功。3.构建成功的双元载体转化入根癌农杆菌中,PCR结果验证转化成功后,用带有双元载体的根癌农杆菌转化犬小孢子菌,通过PCR和Southern blot对转化子进行鉴定,筛选出犬小孢子菌Zrta基因与潮霉素B抗性基因发生同源重组的菌株,获取的Zrta基因缺失株连续3次传代培养后,遗传性状稳定。本研究通过转录组测序明确了锌缺乏对犬小孢子菌基因表达和生物代谢等的影响,其中Zrta基因的基因注释结果表明其在锌代谢中可能起关键作用,成功构建了Zrta基因转化用的双元载体pDHt/ZrtaⅠ、Ⅱ::hph,通过ATMT成功获得犬小孢子菌Zrta基因缺失株,为Zrta基因的致病性和功能验证奠定了基础。
Abstract
