作者(2019)在《电喷雾法制备海藻酸水凝胶微马达及其磁驱动运动研究》一文中研究指出:流体电喷雾法(Hydrodynamic Electrospray Ionization Jetting Method(HESIJ))是一种可用于制备水凝胶微粒的重要方法,具有诸多优点。首先,HESIJ作为电喷雾质谱(ESI-MS)和离子迁移光谱(ESI-IMS)及其联用技术(如ESI-IMS-MS)中所采用的电离方法,已经广泛应用于分析化学领域,具有电离过程温和的特点,不会破坏需电离的待测物(如蛋白质等生物分子)。其次,HESIJ具有实验装置简单和搭建成本低的优点。最后,HESIJ在制备水凝胶颗粒或胶囊方面具有效率高的特点。在电喷雾针上施加电压并启动注射泵输送电喷雾溶液几秒钟后就可以迅速产生大量颗粒或胶囊。本论文通过优化和改进HESIJ方法,以天然高分子海藻酸盐为原料,以二价钙离子或铜离子为交联剂,以电喷雾的方式制备了海藻酸盐水凝胶颗粒和胶囊,获得了优化的HESIJ实验参数,分析了电喷雾过程中的机理。进一步,通过集成磁性纳米颗粒制备了海藻酸微马达,并实现了其磁驱动运动。本论文完成了以下研究工作:首先,针对现有HESIJ方法制备水凝胶微粒研究中的电喷雾模式较少(主要为锥注模式)以及实验参数有待优化等问题,对利用HESIJ方法制备钙交联海藻酸水凝胶微粒的滴落模式和多射流模式进行了系统的研究。研究表明,海藻酸水凝胶微粒在三种电喷雾模式(滴落、锥注和多喷射模式)、四种喷雾距离(5、10、15和20 cm)和六种喷雾浓度下显示出不同的直径和长径比。比较三种电喷雾模式,锥注模式(即双喷射模式)可以产生粒径最小、长径比最低和尺寸参数浮动最小的水凝胶微粒。对于所有电喷雾模式,当被喷雾溶液浓度≥2.5%时,形成的颗粒直径与喷涂距离是无关的。在不同的电喷雾模式和喷涂距离下,微粒的长径比差异明显。当海藻酸钠溶液浓度≤1.5%且喷雾距离为20 cm时,所有电喷雾模式所产生的微粒长径比均逐渐增大。这种现象可用链喷射效应来解释。其次,提出了超声辅助的HESIJ制备海藻酸水凝胶微粒的方法。当向液体表面喷射水凝胶时会产生表面饱和效应,形成一个不断增长的水凝胶层,直至水凝胶微粒的形成过程终止。采用超声波可以有效防止电喷雾制备水凝胶过程中的表面饱和效应。系统研究了喷涂距离、电喷雾模式、海藻酸浓度、超声频率以及对电极形状等参数对水凝胶微纳米颗粒尺寸的影响。通过在电喷雾制备过程中集成生物相容性磁性纳米颗粒成功制备了海藻酸水凝胶微马达。该微马达可以在强度为5 mT到20 mT且频率在1Hz到100 Hz的旋转磁场驱动下进行自驱运动。这些旋转的海藻酸水凝胶微马达可在溶液中进行定向运动,在磁控药物输送方面具有一定的应用潜力。然后,针对在负电位进行电喷雾操作条件下海藻酸钠易形成致密水凝胶颗粒而难以形成空心微胶囊的问题,利用低频超声辅助HESIJ方法实现了生物可降解的钙交联海藻酸水凝胶微胶囊的制备。系统的实验研究表明,海藻酸盐微胶囊的尺寸和形状与喷雾距离、电喷雾模式、电极形状和被喷雾溶液的浓度等因素是密切相关的。通过调控实验条件,成功制备了花瓶状、蘑菇状、球状等海藻酸盐微胶囊,其平均尺寸可在10μm至2 mm的区间内调控。这些微胶囊可以用于负载药物。微胶囊在负载磁性纳米粒子后可转变为海藻酸水凝胶微马达,能够在旋转磁场推动下进行定向运动。
