作者王桂芳(2019)在《非球形荧光α-Fe2O3纳米粒子的制备与细胞成像研究》一文中研究指出:纳米粒子(nanoparticles)作为药物、成像剂和治疗剂的载体是近年来纳米医学领域研究的热点。与常见的球形纳米粒子相比,非球形纳米粒子在纳米载药领域具有显著不同的性质,如更长的血液循环时间、更低的巨噬细胞内吞率或更高的肿瘤细胞摄入率。但是,迄今为止人们对非球形纳米粒子载体的生物性质,特别是纳米粒子形貌对药物输送体系的影响仍缺乏系统的研究,各个独立研究中采用的实验设计和纳米材料性质的差异导致了研究结论的多样性。针对这一问题,本论文采用合成简单、形貌易调控、表面易修饰和生物相容性好的α-Fe2O3作为模型体系,利用经典的水热法制备了纺锤形、椭球形和类球形三种α-Fe2O3纳米粒子,通过后修饰的方法在其表面形成含有具有聚集诱导荧光增强效应的小分子荧光染料的SiO2薄层,得到了两组荧光纳米粒子,细胞成像研究初步揭示了荧光纳米粒子形貌对细胞摄取行为的影响。本论文的主要内容如下:第一章:绪论。在综述纳米粒子载体及其形貌对其生物行为影响研究进展的基础上,提出了本论文的研究课题。第二章:非球形荧光α-Fe2O3纳米粒子(FNP1)的制备与细胞成像研究。采用水热法制备出纺锤形、椭球形和类球形三种形貌的α-Fe2O3纳米粒子,利用聚乙烯吡咯烷酮(PVP)稳定α-Fe2O3纳米粒子,得到二者的复合物α-Fe2O3@PVP。然后将四苯基乙烯功能化的硅氧烷衍生物(AIEgen1)和正硅酸乙酯(TEOS)加入到上述α-Fe2O3@PVP体系中,通过一锅溶胶-凝胶反应制备出三种形貌的FNP1(纺锤形,椭球形和类球形分别记作:SNP1,ENP1和QSNP1)。分别利用透射电镜(TEM)和荧光光谱仪详细考察了AIEgen1和TEOS的含量对SNP1粒径大小和荧光发射光谱的影响。结果表明SiO2壳层厚度随着TEOS的含量增加而增加,通过调节这些组分的浓度,可以将SiO2壳层厚度控制在26-54 nm范围内。随着AIEgen1或TEOS的含量的增加,所制备的荧光纳米粒子荧光越强,荧光量子产率越高。动态光散射(DLS)实验结果表明纺锤形、椭球形和类球形纳米粒子的Zeta电位分别是-24.3 mV,-26.1 mV和-29.6 mV,表明FNP2纳米粒子表面带有负电荷。此外,由于FNP1具有较低的细胞毒性和良好的光稳定性,利用共聚焦荧光显微镜研究了FNP1的形貌对其细胞摄取行为的影响。初步结果表明:纺锤形纳米粒子与椭球形纳米粒子进入细胞的速率相近,而细胞对类球形的内吞速率低于纺锤形和椭球形。第三章:非球形荧光α-Fe2O3纳米粒子(FNP2)的制备与细胞成像研究。与第二章方法类似,采用另一种小分子荧光染料四苯基乙烯功能化的硅氧烷衍生物(AIEgen2)和TEOS在三种形貌的α-Fe2O3@PVP表面上进行溶胶-凝胶反应,得到三种形貌的FNP2(纺锤形,椭球形和类球形分别记作:SNP2,ENP2和QSNP2)。利用TEM探究了AIEgen2和TEOS的含量对SNP2粒径大小的影响,结果表明随着反应体系中AIEgen2含量的增加逐渐出现SiO2自成球的现象。而随着TEOS浓度增大,SiO2厚度逐渐增加。利用荧光光谱仪探究了TEOS的含量对SNP2荧光光谱的影响,结果表明TEOS的含量越多,制备的纳米粒子的荧光越强,荧光量子产率越高。动态光散射(DLS)实验结果表明纺锤形、椭球形和类球形纳米粒子的Zeta电位分别是+48.2 mV,+51.3 mV和+50.5mV,表明FNP2纳米粒子表面带有正电荷。此外,初步探究了不同形貌FNP2在HeLa细胞中的荧光成像。第四章:全文总结。
