作者吴江萍(2019)在《植物乳杆菌CCFM8610缓解镉暴露诱导肠道损伤的效果评价及机制探究》一文中研究指出:镉(Cadmium,Cd)是一种在环境中广泛存在的重金属,近年来工业化进展的不断加速及污水农田灌溉技术的广泛使用,镉污染事件频频发生,严重危害了公共健康。过往研究表明,植物乳杆菌CCFM8610是一株对重金属镉具有良好的吸附能力的乳酸菌,可有效缓解机体内炎症,预防日常镉暴露。但现阶段,关于植物乳杆菌CCFM8610缓解镉暴露的研究主要集中在肝、肾方面。而肠道作为镉吸收的直接器官,植物乳杆菌CCFM8610对肠道镉暴露损伤的恢复效果及机制却并未有靶向研究。因此,本课题通过体内外实验,对植物乳杆菌CCFM8610恢复镉暴露诱导肠道损伤的效果及其机制进行了探究。首先通过构建慢性镉暴露动物模型,探究植物乳杆菌CCFM8610对慢性镉暴露小鼠肠道损伤的恢复能力。结果表明植物乳杆菌CCFM8610能有效降低小鼠脏器内约20%的镉蓄积,同时加快慢性镉暴露小鼠排便速度,缩短镉暴露小鼠排便时间近2小时,增强小鼠结肠肌条张力。检测小鼠肠道中神经递质,并对小鼠结肠c-kit蛋白进行免疫标记后发现,植物乳杆菌CCFM8610显著调节了镉暴露紊乱的肠道神经递质一氧化氮、5羟色胺、P物质、乙酰胆碱酯酶、血管活性肠肽和降钙素基因相关肽的含量,保护了肠道Cajal细胞,并对肠道运动能力有一定恢复效果。证明植物乳杆菌CCFM8610干预后,能有效预防镉暴露诱导的肠道便秘症状。进一步通过对肠道中JNK、ERK、bcl-2以及膜蛋白的分析发现,植物乳杆菌CCFM8610抑制了肠道ERK和JNK的表达,提高了bcl-2相对表达,促进了肠道细胞的抗凋亡活动,缓解了镉暴露造成的肠道应激与细胞凋亡。同时植物乳杆菌CCFM8610也显著提高了肠道上皮细胞中Claudin、DMT-1和E-cadherin蛋白的相对表达,保护了肠道上皮细胞膜蛋白和肠道屏障。此外,对小鼠肠道菌群的16S测序发现,植物乳杆菌CCFM8610也具有调节肠道菌群,保护肠道微环境的作用。证明植物乳杆菌CCFM8610能通过保护肠道屏障、抑制肠道内炎症和保护肠道菌群稳态这三个途径来保护肠道免受镉暴露造成的损伤。通过构建镉暴露肠上皮(Human colon tumor cell line,HT-29)细胞模型,以蛋白组学手段检测到了4089个可信蛋白,筛选得到168个差异蛋白。基于Gene Ontology数据库、KEGG蛋白注释及蛋白互作分析发现,重金属镉引起了细胞内的压力应激,产生了大量丝裂原活化蛋白激酶(Mitogen-activated protein kinase,MAPK)、核因子-κB(Nuclear factor kappa-light-chain-enhancer of activated B cells,NF-κB)通路相关蛋白,并扰乱了上皮细胞膜上FLAN等蛋白的含量,导致细胞膜结构损坏。使用植物乳杆菌CCFM8610干预细胞后发现,植物乳杆菌CCFM8610有效抑制了镉暴露激活的Ras-Raf-MEK-ERK、NF-κB信号通路,从而缓解了细胞内压力应激和细胞内炎症。此外,植物乳杆菌CCFM8610也降低了镉暴露细胞内蛋白AKR1B10相对表达量,提高了蛋白DDX21和蛋白FLNA的相对表达,从而调节了细胞内能量代谢、RNA和蛋白合成,并通过保护细胞膜蛋白Claudin-1和E-cadherin,维持细胞结构,保护细胞屏障。通过与该实验中其他菌株的对比发现,植物乳杆菌CCFM8610可以通过特意性调节蛋白AKR1B10、DDX21和FLNA的相对表达,从而抑制细胞内炎症保护细胞屏障。与植物乳杆菌CCFM175、CCFM408相比对有一定的菌株特异性。
