作者倪湾(2019)在《洋葱槲皮素抗炎活性及对NF-κB和Akt信号通路的影响》一文中研究指出:洋葱属百合科葱属二年生草本植物,因富含槲皮素、前列腺素A等重要生物活性物质,而成为具有多种营养价值的蔬菜。洋葱槲皮素(onion quercetin,OQE)是洋葱中提取的一种多羟基黄酮类化合物,现代研究表明,洋葱槲皮素具有抗炎、抗氧化、抗癌、降糖、降压、免疫调节及保护心血管等功效。为探讨洋葱槲皮素的抗炎作用,本文采用1μg/mL脂多糖处理小鼠腹腔巨噬细胞建立细胞炎症模型,体外研究洋葱槲皮素的抗炎活性及其作用机制,为开发抗炎功能性食品提供实验依据。本文的研究方法和结果如下:1.体外建立LPS诱导的体外小鼠腹腔巨噬细胞炎症模型,采用MTT法检测OQE单药组、OQE与LPS复合药组对小鼠腹腔巨噬细胞吞噬功能的影响;结果表明1μg/mL LPS作用小鼠腹腔巨噬细胞24 h,在20~160μg/mL剂量范围内,OQE单药组刺激、OQE与LPS复合药组均能提高小鼠腹腔巨噬细胞的吞噬能力。2.采用ELISA法检测OQE对LPS诱导的小鼠腹腔巨噬细胞分泌细胞因子及炎症介质IL-1β、COX-2、IL-6、和TNF-α的影响;结果表明1μg/mL LPS作用小鼠腹腔巨噬细胞24 h,OQE处理组均可抑制LPS诱导的小鼠腹腔巨噬细胞IL-1β和COX-2、IL-6和TNF-α的分泌,且随着OQE浓度增加,抑制作用增强,存在着明显的剂量-效应关系。3.采用RT-PCR法检测OQE对LPS诱导的小鼠腹腔巨噬细胞分泌细胞因子及炎症介质基因IL-1β、COX-2、IL-6和TNF-α的mRNA表达水平;结果表明1μg/mL LPS作用小鼠腹腔巨噬细胞24 h,OQE处理组均可抑制LPS诱导的小鼠腹腔巨噬细胞分泌细胞因子及炎症介质基因IL-1β、COX-2、IL-6和TNF-α的mRNA表达水平,且随着OQE浓度增加,抑制作用增强,存在着明显的剂量-效应关系;OQE可抑制LPS诱导的小鼠腹腔巨噬细胞炎症反应,具有抗炎作用。4.采用Western Blot法检测OQE对LPS诱导的小鼠腹腔巨噬细胞中NF-κB和Akt信号通路蛋白磷酸化水平表达的影响;结果表明1μg/mL LPS作用小鼠腹腔巨噬细胞24h,LPS刺激可以显著上调小鼠腹腔巨噬细胞中NF-κB-p65和Akt蛋白磷酸化水平,而OQE处理组均可抑制LPS诱导的NF-κB-p65和Akt蛋白磷酸化水平;通过对NF-κB和Akt信号通路的研究,发现OQE可抑制NF-κB和Akt信号转导通路进而抑制炎症因子的表达,从而发挥抗炎作用。

Abstract
