作者官艳东(2019)在《棕榈酸对围产期奶牛中性粒细胞胞外诱捕网形成的影响》一文中研究指出:围产期奶牛在产后体内易出现能量负平衡,从而使得血浆中非脂化脂肪酸的浓度大大升高。最新的研究证实高浓度的非脂化脂肪酸可以导致围产期亚急性炎症尤其是代谢性炎症。同时,棕榈酸是非脂化脂肪酸毒性最大且变化最大的脂肪酸,有可能参与炎症与免疫抑制,但进一步的机制并不明确。中性粒细胞作为动物机体先天免疫系统的主要效应细胞,较新的机制发现中性粒细胞胞外诱捕网的形成是免疫机制之一。为明确中性粒细胞胞外诱捕网的形成是否为棕榈酸诱导炎症和免疫应答的机制,本研究将探讨奶牛围产期对中性粒细胞胞外诱捕网形成的影响和机制。研究采用红细胞裂解法分离了围产期产后奶牛(DIM:7-12)的外周血中性粒细胞培养,使用荧光显微镜(SYTOX Orange染料)、扫描电子显微镜观察中性粒细胞胞外诱捕网的形态,使用Picogreen荧光染料对双链DNA进行定量测定,TMB法测定髓过氧化物酶(MPO)的水平,来明确棕榈酸是否能引起中性粒细胞胞外诱捕网的形成。其次,使用比色法检测乳酸脱氢酶活性,使用DHCF法检测活性氧水平,辣根过氧化物酶比色法检测过氧化氢的活性水平,Western blot技术检测PI3K/Akt通路相关蛋白Akt、p-Akt、NE、Nox2和H3的表达水平,探讨棕榈酸诱导的中性粒细胞胞外诱捕网的机制。结果发现:分离得到中性粒细胞的平均活率为97%,纯度为96%。相对于对照组,棕榈酸可导致乳酸脱氢酶活力显著降低,细胞外DNA的释放显著增加,细胞外双链DNA含量显著增多,胞外网状结构数量显著增多。棕榈酸可引起中性粒细胞胞内ROS和H2O2水平显著升高,但L-Epicatechin使得ROS和H2O2水平显著降低。棕榈酸可导致中性粒细胞的MPO活性水平显著上升,L-Epicatechin可使抑制棕榈酸诱导的中性粒细胞胞外MPO水平。棕榈酸可引起Nox2,Akt和p-Akt蛋白表达水平均显著增加,L-Epicatechin可抑制棕榈酸诱导的Nox2,Akt和p-Akt蛋白表达水平显著增加。结论:棕榈酸可以导致中性粒细胞胞外诱捕网的生成;其机制可能是激活PI3K/Akt通路相关蛋白,引起中性粒细胞产生氧化应激,导致中性粒细胞胞外诱捕网的形成。
