作者邬蒙(2019)在《红皮梨着色相关光响应基因的筛选与功能验证》一文中研究指出:红皮梨果实颜色形成是一个复杂的过程,光是影响果皮颜色形成的重要环境因素,但是梨中调控花青苷合成相关的光响应基因知之甚少。因此,研究光响应基因调控红皮梨果皮花青苷合成与降解的机理尤为重要。本课题首先采用了六个不同红皮梨品种分析梨果皮花青苷着色模式的差异。然后通过‘早酥’梨高温高光下果皮变红的转录组筛选出调控花青苷合成的光响应基因PbCOP1.1、PbPIF3.1和PbZAT12。研究表明,PbCOP1.1负调控花青苷积累,PbPIF3.1和PbZAT12正调控花青苷积累。主要研究结果如下:1.不同红皮梨品种着色模式的差异。六个红皮梨品种(‘5号优系’、‘红巴梨’,‘红星’、‘红丝绸’、‘红早酥’、‘红佩雷莎’)花青苷分析结果显示,花青苷积累高峰均发生在果实发育的中期(花后55天和75天),花青苷含量先上升后下降,后期出现果实褪色的情况。荧光定量分析结果显示,花青苷合成结构基因F3H和UFGT2的是六个品种花青苷合成的关键基因,与花青苷含量呈现出较高的正相关性。此外,花青苷合成转录因子MYB10和bHLH3也是花青苷积累的关键基因。根据基因表达水平所做的主成分分析和层次聚类分析结果显示,六个品种被分为三种不同花青苷积累模式,‘红早酥’(东方梨)分为一组,由基因PAL、ANS1、CHS、DFR1、bHLH33、MYB10b和PIF3.1决定;‘红丝绸’(西洋梨)和‘红星’(西洋梨)被分为一组,由基因UFGT1、MYB10、COP1、和ANR决定;‘红佩雷莎’(西洋梨)‘红巴梨’(西洋梨)和‘5号优系’(西洋梨×东方梨)分为一组,由这些基因共同决定。2.PbCOP1.1负调控花青苷积累。从‘红早酥’中克隆得到PbCOP1s。组织特异性表达分析及叶片不同发育时期定量结果显示,PbCOP1s在叶片中大量表达,而且在绿色叶片中的表达量显著高于红色叶片中的表达量。在‘红巴梨’果皮过表达PbCOP1.1和PbCOP1.2后,与对照相比,梨果皮注射区中PbCOP1.1表达量显著升高,花青苷含量显著减少;PbCOP1.2则无明显变化。定量分析过表达PbCOP1.1后花青苷合成相关基因的表达水平,结果显示CHI、DFR、UFGT2、bHLH3、HY5和GST表达量显著下调。3.PbPIF3.1和PbZAT12正调控花青苷的合成。在‘早酥’果皮中过表达PbPIF3.1和PbZAT12能促进花青苷积累,在‘红星’果皮中沉默PbPIF3.1和PbZAT12能减少花青苷的积累。亚细胞定位结果显示,PbPIF3.1和PbZAT12蛋白定位于细胞核。过表达PbPIF3.1后定量结果显示,花青苷合成相关基因都显著上调了,除了MYB10b。此外,PbZAT12的表达水平也显著上调。沉默PbPIF3.1后,花青苷合成的相关基因中,仅UFGT2、MYB10和GST显著下调。双荧光素酶活性分析显示,PbPIF3.1可以激活F3H启动子的活性。过表达PbZAT12后,CHS、DFR1、ANS1和UFGT2都呈现出显著上调,其他基因无显著差异。沉默PbZAT12后,花青苷合成相关基因表达水平下调,PAL、UFGT2、MYB10、MYB10b、bHLH3和bHLH33显著下调。
