作者邓强(2019)在《扁鹊名方“三豆饮”饮料的研制》一文中研究指出:目的:在古代著名医家扁鹊名方三豆饮(黑豆、绿豆、赤小豆)的基础上,研制具有清热解毒、活血祛风、养肝润肺、润燥生津作用,口感良好、易为大众接受、服用方便的三豆饮饮料。方法:1三豆饮饮料的配制(1)建立有效物质含量测定方法:采用苯酚-硫酸法,测定三豆饮中总多糖含量,采用Folin-Ciocalteu法测定三豆饮中总多酚含量。(2)优化三豆饮水提工艺:在单因素试验基础上,利用响应面法以提取次数、料液比、提取时间为因素,总多糖、总多酚提取量为响应值,优化三豆饮提取工艺。(3)比较产物抗氧化性:用DPPH法和羟自由基法测定三豆饮饮料原提取液和优化提取液的抗氧化作用。(4)确定三豆饮调配工艺:通过感官评价确定三豆饮饮料的配料,以微生物指标、褐变指数、功效指标为标准,确定灭菌条件及护色条件。(5)确定三豆饮保质期:在5℃、25℃、37℃储藏条件下,90天储藏期中,测定三豆饮护色组饮料和三豆饮未护色组饮料中微生物指标、理化指标和功效指标变化情况。2豆渣膳食纤维提取工艺研究。在酸提单因素实验基础上,采用响应面法以提料液比、酸提时间、酸提温度、酸提pH为因素,渣膳食纤维含量为响应值,以获得适宜酸提工艺;在最佳酸提工艺条件下,在碱提单因素实验基础上,采用响应面法以碱提料液比、碱提时间、碱提温度、碱提pH为因素,渣膳食纤维含量为响应值,以获得适宜豆渣膳食纤维碱提工艺。3三豆饮饮料质量标准研究根据三豆饮饮料调味工艺、杀菌工艺和护色工艺确定调配工艺标准,根据保质期中物生物指标,理化指标和功效指标变化情况确定三豆饮饮料保质期和各指标变化标准。结果:1三豆饮饮料的配制(1)D-无水葡萄糖对照品在30.1 μg/mL~1 0lμg/mL范围内呈良好线性关系(2)(r=0.9992),平均回收率为97.24%,RSD为1.74%(n=9)。没食子酸对照品在23μg/m L~69 μg/m L范围内呈良好线性关系(r=0.9992),平均回收率为97.28%,RSD 为 1.47%(n=9)。(2)三豆饮最佳水提工艺条件为:提取次数3次,料液比1:18(g:mL),提取时间2.5 h,在此条件下三豆饮提取液中总多糖含量为242.9 mg/g、总多酚的含量为 8.86 mg/g。(3)原水提取液DPPH.清除率为72.9%,羟自由基清除率为95%。适宜提取溶液DPPH率为74.5%,羟自由基清除率为97%。最佳提取溶液清除DPPH-IC50的质量浓度为206.2 μg/mL、清除羟自由基IC50的质量浓度为184μg/mL;总多糖浓度在 70.5 μg/m L-1127.5 μg/mL,总多酚浓度 2.6 μg/m L-40.5 μg/m L 浓度范围时,样品溶液的抗氧化性随总多糖和总多酚含量的增加而增加。(4)通过感官评价确定三豆饮饮料配料为:三豆饮提取液100 mL、白砂糖2000 mg、盐75 mg。适宜灭菌条件为:灭菌温度110℃,时间1min。最佳护色工艺为:0.3%的D-异抗坏血酸钠。(5)90天储藏过程中发现在5℃储藏条件下,护色组与未护色组微生物指标、理化指标、功效指标基本保持稳定。25℃储藏条件下,护色组各指标基本稳定;未护色组80天有菌落生成,功效指标有所降低。37℃储藏条件下,护色组与未护色组60天后有菌落生成,理化指标和功效指标有所降低,未护色组各指标下降速度均大于护色组。2豆渣膳食纤维提取工艺研究。三豆饮豆渣膳食纤维最佳酸提条件为:料液比(1:25)、酸提时间1.5 h、酸提温度87℃、酸提pH4.65。酸提后膳食纤维为59%,比原豆渣膳食纤维含量增加14.4%。最佳碱提条件为:料液比(1:35)、碱提时间4h、碱提温度56℃、碱pH11.8。碱提后膳食纤维为75.7%,碱提后膳食纤维含量比原豆渣膳食纤维含量增加30.1%。3三豆饮饮料各检查项均符合相关规定。结论:1三豆饮饮料的配制(1)所建立的分析方法简便、准确、可靠,重复性好;(2)三豆饮提取液总多糖、总多酚含量实测值和预测值基本一致,该工艺稳定可行;(3)原提取液与最佳提取液具有较好抗氧化活性;(4)三豆饮饮料调配工艺稳定可行;(5)三豆饮饮料25℃保质期为3个月,保质期较长。2豆渣膳食纤维提取工艺研究。豆渣中膳食纤维含量实测值和预测值基本一致,该工艺稳定可行。3拟定的三豆饮饮料质量标准有效、稳定、可控。

1.47%(n=9)。(2)san dou yin zui jia shui di gong yi tiao jian wei :di qu ci shu 3ci ,liao ye bi 1:18(g:mL),di qu shi jian 2.5 h,zai ci tiao jian xia san dou yin di qu ye zhong zong duo tang han liang wei 242.9 mg/g、zong duo fen de han liang wei 8.86 mg/g。(3)yuan shui di qu ye DPPH.qing chu lv wei 72.9%,qiang zi you ji qing chu lv wei 95%。kuo yi di qu rong ye DPPHlv wei 74.5%,qiang zi you ji qing chu lv wei 97%。zui jia di qu rong ye qing chu DPPH-IC50de zhi liang nong du wei 206.2 μg/mL、qing chu qiang zi you ji IC50de zhi liang nong du wei 184μg/mL;zong duo tang nong du zai 70.5 μg/m L-1127.5 μg/mL,zong duo fen nong du 2.6 μg/m L-40.5 μg/m L nong du fan wei shi ,yang pin rong ye de kang yang hua xing sui zong duo tang he zong duo fen han liang de zeng jia er zeng jia 。(4)tong guo gan guan 59%,bi yuan dou zha shan shi qian wei han liang zeng jia 14.4%。zui jia jian di tiao jian wei :liao ye bi (1:35)、jian di shi jian 4h、jian di wen du 56℃、jian pH11.8。jian di hou shan shi qian wei wei 75.7%,jian di hou shan shi qian wei han liang bi yuan dou zha shan shi qian wei han liang zeng jia 30.1%。3san dou yin yin liao ge jian cha xiang jun fu ge xiang guan gui ding 。jie lun :1san dou yin yin liao de pei zhi (1)suo jian li de fen xi fang fa jian bian 、zhun que 、ke kao ,chong fu xing hao ;(2)san dou yin di qu ye zong duo tang 、zong duo fen han liang shi ce zhi he yu ce zhi ji ben yi zhi ,gai gong yi wen ding ke hang ;(3)yuan di qu ye yu zui jia di qu ye ju you jiao hao kang yang hua huo xing ;(4)san dou yin yin liao diao pei gong yi wen ding ke hang ;(5)san dou yin yin liao 25℃bao zhi ji wei 3ge yue ,bao zhi ji jiao chang 。2dou zha shan shi qian wei di qu gong yi yan jiu 。dou zha zhong shan shi qian wei han liang shi ce zhi he yu ce zhi ji ben yi zhi ,gai gong yi wen ding ke hang 。3ni ding de san dou yin yin liao zhi liang biao zhun you xiao 、wen ding 、ke kong 。
