作者肖斌(2019)在《公鼠束缚应激诱发精子和生精细胞凋亡:Fas/FasL系统通过促进附睾TNF-α和TRAIL分泌引起精子凋亡》一文中研究指出:现代社会充满了压力,压力是一种严重的心理应激,心理应激会造成不孕不育。因此人们的生殖健康正面临巨大的挑战。应激对雄性生殖能力的损伤可能通过破坏生殖内分泌和造成细胞凋亡等方式进行,破坏精子发生过程,损伤精子质量,进而降低精液质量,同时关于应激对成熟精子影响的研究较少。在很大程度上,心理应激损伤精液质量的机制尚不明确。因为束缚应激模型可以较好地模拟人类日常生活压力所造成的心理应激。所以本研究中我们用小鼠束缚应激模型模拟心理应激研究了束缚应激成年公鼠导致各类生精细胞和精子凋亡的情况及其与Fas/FasL系统的关系。我们对昆明白公鼠束缚应激48小时后,检测其精子的活力、精子和生精细胞的凋亡情况及精子和生精细胞内活化Caspase-3和Fas蛋白的表达情况、附睾尾中活化Caspase-3和Fas/FasL蛋白的表达情况,同时用FasL功能缺失型突变的gld品系小鼠和gld的对照鼠—野生型C57BL/6J品系小鼠进行相关实验,并在体外添加sFasL保存精子后检测精子活力和凋亡,证实了Fas/FasL信号系统在束缚应激中对精子和生精细胞的促凋亡作用,阐明了该系统在束缚应激过程中促进附睾尾中成熟精子凋亡的机制。实验结果如下:(1)束缚应激导致公鼠血清皮质醇水平显著升高,在束缚应激开始后1小时、24小时和48小时,小鼠皮质醇水平始终显著高于对照组,说明我们的应激模型可以有效造成公鼠应激;(2)束缚应激组与对照组精子密度没有显著差异;(3)束缚应激组与对照组新取出的精子活力没有显著差异,但自体外保存90分钟起,束缚组精子活力显著低于对照组,说明束缚应激加快了体外条件下精子活力下降的速度;(4)束缚应激导致Annexin-V FITC阳性精子比例显著升高,精子中活化Caspase-3和Fas蛋白含量显著升高,说明束缚应激48小时促进精子凋亡,且束缚应激对精子的促凋亡作用可能与Fas/FasL系统有关;(5)束缚应激48小时导致附睾尾活化Caspase-3和Fas/FasL蛋白表达水平显著升高,说明束缚应激会促进附睾凋亡;(6)束缚应激48小时导致Annexin阳性生精细胞比例显著升高,生精细胞中活化Caspase-3蛋白表达量显著升高,说明束缚应激48小时促进生精细胞凋亡;束缚应激后第35天束缚组精子活力显著低于对照组,凋亡比例显著高于对照组,说明束缚应激导致生精细胞凋亡进而影响其分化而来的精子的质量;(7)对照组gld和C57BL/6J小鼠精子的体外受精率、凋亡比例,生精细胞凋亡比例均无显著差异,束缚应激48小时后,gld和C57BL/6J小鼠上述指标均发生显著不良变化,但gld小鼠各指标受影响的程度显著小于C57BL/6J小鼠,这说明Fas/FasL系统参与束缚应激导致的精子和生精细胞质量损伤,但也可能存在其他通路在这一过程中发挥作用;(8)精子体外保存过程中添加sFasL并不影响精子活力和凋亡,而sTNF-α显著降低精子活力并升高其凋亡比例,对照组gld和C57BL/6J小鼠附睾尾TNF-α和TRAIL表达量无显著差异,束缚应激48小时后,附睾尾中FasL蛋白表达显著上升,gld和C57BL/6J小鼠附睾尾TNF-α和TRAIL表达量均显著上升,但gld小鼠TNF-α和TRAIL表达量均显著低于C57BL/6J小鼠,这说明束缚应激上调附睾尾中FasL表达促进TNF-α和TRAIL分泌间接导致精子凋亡,但也可能存在独立于Fas/FasL系统之外的通路(TNF-α/TNFR1或TRAIL/TRAILR)在其中发挥作用,且作用强于Fas/FasL系统。因此,我们的结果表明:束缚应激模型能有效造成公鼠应激,束缚应激通过激活Fas/FasL系统促进成熟精子凋亡,并通过导致生精细胞凋亡产生低质量精子,在附睾中FasL因束缚应激而表达升高,促进TNF-α和TRAIL的产生,间接导致精子凋亡。
Abstract
