作者段明亮(2019)在《PJPs结构域改变作为离心运动引起的骨骼肌过度使用损伤起因因素的研究》一文中研究指出:1目的:过度使用性损伤是一种常见的骨骼-肌肉系统疾病,具有极强的运动及工作相关性,对竞技运动员及普通劳动人群的生活产生了严重的影响。过度使用性损伤极为常见,但其发生机制尚未明确。以国内外学者的研究为依据,发现骨骼肌过度使用性损伤可能与骨骼肌肌束膜衔接盘(perimysial junctional plates,PJPs)结构域的变化关系密切。故本研究采用离心训练,从而引起过度使用性损伤,并基于PJPs结构域的变化,探讨造成骨骼肌过度使用性损伤的机制。2方法:30只健康雄性Wistar大鼠经一周适应性训练后,随机分为安静组(CON)、4周连续训练组(4E)、4周隔天训练组(4N)、4周连续训练1周自然恢复组(1RG)以及4周隔天训练1周自然恢复组(1RNG)5个组别。各组正常饮食,CON组自由活动,4E组进行为期4周的训练(跑台坡度-16°,速度1620m·min-1,6090min·d-1,5天/周),4N组进行为期4周的训练(跑台坡度-16°,速度1620m·min-1,6090min·d-1,3天/周),1RG与1RNG组于训练结束后自然恢复一周。CON组在4周结束后即刻进行取材(趾长伸肌);4E组与4N组在末次训练后36h后进行取材;1RG组与1RNG组在第5周结束后即刻进行取材。透射电子显微镜观察PJPs结构域变化;扫描电子显微镜(1.5 mol/L NaOH消蚀和非消蚀法)观察肌束膜胶原纤维网络分枝变化。分析各组大鼠血液指标变化。3结果:3.1 4E组PJPs结构域变化:(1)肌膜外PJPs结构中骨骼肌肌束膜胶原纤维网络分支缆及其细小分支增多,PJPs数量呈现过度减少的状态(P<0.001);(2)肌膜PJPs结构中胞膜窖数量增多(P<0.001),肌膜重度降解(P<0.001);(3)肌膜内PJPs结构中肌膜下线粒体出现线粒体肿胀,肌原纤维间线粒体空泡;肌节严重缩短(P<0.001)、肌节结构紊乱。3.2 1RG组PJPs结构域变化:(1)肌膜外PJPs结构中骨骼肌肌束膜胶原纤维网络分支缆及其细小分支增多,PJPs数量呈现过度增加的状态(P<0.001);(2)肌膜PJPs结构中胞膜窖数量增加(P<0.001),略少于4E组,肌膜降解程度依旧严重但较4E组降低(P<0.001);(3)肌膜内PJPs结构中肌膜下线粒体与肌原纤维间线粒体可见明显线粒体融合现象,肌节损伤严重程度降低,肌节长度仍明显缩短(P<0.001)。3.3 4N组PJPs结构域变化:(1)肌膜外PJPs结构中骨骼肌肌束膜胶原纤维网络分支缆及其细小分支增多,PJPs数量呈现增加的状态(P<0.001),但幅度较小;(2)肌膜PJPs结构中膜窖数量增多(居5个组别之首)(P<0.001),大小不均,肌膜中度降解;(3)肌膜内PJPs结构中肌膜下线粒体与肌原纤维间线粒体存在线粒体融合现象,肌节严重缩短(仅次于4E组)(P<0.001),肌节结构紊乱。3.4 1RNG组PJPs结构变化:(1)肌膜外PJPs结构中骨骼肌肌束膜胶原纤维网络分支缆及其细小分支减少,PJPs数量基本恢复;(2)肌膜PJPs结构中胞膜窖数量基本恢复,肌膜轻度降解;(3)肌膜内PJPs结构中肌膜下线粒体与肌原纤维间线粒体结构基本正常,肌节长度与结构基本恢复。3.5各组大鼠血液指标变化:4E、4N、1RG、1RNG组血清H2O2含量、MDA含量、LDH含量均较CON组增加,且1RG组H2O2含量、MDA含量、LDH含量增加高于4E组;1RNG组H2O2含量、MDH含量高于4N组,1RG组与1RNG组LDH含量无显著性差异。4结论:4.1通过观察骨骼肌PJPs结构域变化发现,隔天离心训练组效果优于连续离心训练组。4.2过度离心训练使肌束膜胶原纤维网络分支缆细小分支减少,与肌内膜、肌膜发生偏离,影响肌束膜衔接盘结构域稳态,造成过度使用性损伤的出现。4.3过度离心训练1周自然恢复后,PJPs数量增加,影响肌束膜衔接盘结构域稳态恢复,骨骼肌损伤修复缓慢。4.4过度离心训练造成的PJPs结构域稳态改变与运动期间H2O2含量变化有关,导致骨骼肌损伤修复时间延长。
Abstract
