作者贾杨彦圣(2019)在《探究Slc5a8在小鼠眼角膜上皮表达的时空分布》一文中研究指出:角膜缘干细胞(Limbal stem cells,LSCs)存在于角膜缘区域。在个体发育过程中,LSCs向心移行并分化为成熟角膜上皮细胞。据此,可以作为LSCs和成熟角膜上皮细胞的鉴定和定位的理论基础。小鼠是重要的模式动物之一,但关于小鼠角膜缘细胞基因差异性表达的研究尚未见相关报道。本课题结合分子生物学技术和高通量组学技术探究小鼠眼角膜不同区域的差异表达,加深人们对于小鼠眼角膜细胞基因差异性表达的认知。SLC5A8是钠偶联的单羧酸转运蛋白(也称为SMCT1)参与乳酸和短链脂肪酸的运载,这一运载体不仅可以在卵细胞中表达,在其他体细胞中也同样表达。我们推测在小鼠角膜也会表达。本研究利用HE染色、免疫组化染色进行小鼠眼角膜形态学观察,界定角膜缘区域。在此基础上,通过获取角膜缘组织并将角膜缘区域根据冠状面象限分为上(Superior,S)、下(Inferior,I)、鼻(Nasal,N)、颞(Temporal,T)四个组,以中央角膜区域和结膜区域作为对照组进行转录组测序(RNA-seq)。然后对测序结果进行差异分析、GO注释和KEGG通路分析。通过差异分析获得细胞基因差异性表达的概况,筛选出有研究意义的差异基因并探究其在小鼠眼角膜中不同水平的差异性表达。我们成功地对小鼠角膜缘区域进行了准确界定。18个样品的RNA-seq测序质量均符合公认的要求(Q20≥95%、Q30≥85%),其中,Q20最小值为95.79%,Q30最小值88.71%,clean reads ratio最小值为94.89%。差异分析结果显示Slc5a8在各实验组中差异表达,因此,进一步通过qRT-PCR和Western Blotting对其进行定量和半定量检测,并利用单分子RNA原位检测和免疫组化分别探究其在转录水平和蛋白水平的空间分布情况。qRT-PCR和单分子RNA原位检测结果显示Slc5a8在转录水平主要高表达于角膜缘上皮区域,Western Blotting和免疫组化结果显示SLC5A8在蛋白水平却主要高表达于中央角膜上皮区域。这种表达反差提示Slc5a8基因在转录水平可作为LSCs候选标志物,在蛋白水平可作为角膜上皮细胞候选标志物。
