作者王洁丽(2019)在《驯化对大豆根系微生物群落组装和根瘤内生菌多样性影响的研究》一文中研究指出:植物驯化和育种是农业产生和发展的基础,对人类社会文明产生巨大影响。现代品种的狭窄遗传背景正逐渐成为作物改良的主要瓶颈,而近缘野生种类为作物的驯化和育种提供了机遇。根际微生物已被证明可赋予植物新的抗逆性与生态功能。然而,植物驯化是否影响根际微生物群落的组装和生态功能还不清楚。本文以野生、半野生、栽培和广适种大豆根系微域为研究对象,通过盆栽试验,分别将这些大豆品种种植于来自东北、黄淮海和南方地区的土壤中,再通过16S rDNA高通量测序技术,研究不同大豆根区、根际、根内、根瘤内等四个部位微生物群落的结构与组成,分析不同大豆品种之间根际微生物群落的差异,探究了不同大豆品种对根瘤内生菌群落的影响。得到以下结果:(1)大豆根系微生物群落主要受土壤、根系部位和大豆品种的影响。东北、黄淮海和南方地区的大豆根系微生物群落具有显著差异。从根外到根内,微生物的丰富度和多样性逐渐降低,差异OTUs逐渐增多,根系的过滤作用逐渐增强,且不同大豆品种根系微域对OTUs具有各自的选择过滤作用。通过网络分析发现,野生大豆根内细菌网络结构更复杂,半野生、栽培和广适种大豆根际细菌网络更复杂,根内细菌群落易受大豆种类影响,并且根区、根际和根内的关键物种不同。(2)不同土壤中大豆根际微生物群落结构具有显著差异。从野生、半野生到栽培再到广适种大豆,在东北土壤中,根际细菌丰富度和多样性沿此顺序逐渐降低,然而在黄淮海和南方土壤中,丰富度和多样性变化与之相反;不同大豆品种的核心OTUs数目所占比例较小,但其相对丰度较大。不同大豆品种共同拥有的核心类群,包括Ensifer、Bradyrhizobium、Mesorhizobium、Rhizobium、Pseudomonas、Novosphingobium、Sphingomonas、Massilia、Piscinibacter、Pseudoduganella、Ramlibacter、Variovorax和2个未分类的OTUs。大豆生物表型与不同大豆品种的根际微生物群落显著相关,在OTU水平上,植株全长(株高与根长之和)与野生大豆根际微生物丰度显著正相关,鲜重与栽培大豆根际微生物丰度显著负相关。(3)不同大豆品种之间的根瘤内生菌群落差异显著,且根瘤内生菌群落受土壤类型影响大,其次是不同大豆品种。根瘤内生菌沿着野生、半野生、栽培和广适种大豆的顺序,丰富度和多样性呈逐渐增高。根据根瘤功能特点,我们又将根瘤内生菌分为共生根瘤菌群落和非共生菌群落,结果发现在共生根瘤菌群落中Bradyrhizobium在东北和南方土壤中占主导地位,Ensifer在黄淮海地区占主导地位。在非共生内生菌群落中,慢生根瘤菌科、丛毛单胞菌科、鞘酯菌科、根瘤菌科和肠杆菌科等为主要类群。本研究揭示了野生、半野生、栽培和广适种大豆根系微生物群落组装规律及影响其根瘤内生菌群落多样性的因素,为大豆有益菌群的恢复和利用奠定基础,也为未来育种提供新的思路。
