作者张伟(2019)在《海带降解菌的筛选与发酵产物的成分分析及在植物生长中的应用研究》一文中研究指出:随着海洋活性物质研究的不断发展,人们对海藻多糖的关注日益加强。天然海藻酸不仅具有很好的生物活性,而且其降解产物——海藻寡糖同样具有较强的生物活性,具有很大的开发价值和较为广阔的发展前景。本文主要研究海带降解功能菌的分离、筛选和它们对海带的降解功能,海带发酵产物的成分分析,海带发酵液对油菜种子生长发育的影响及其对土壤微生物菌群的影响。(1)海带降解功能菌的分离、鉴定及筛选本文从山东荣成某海带养殖场采集新鲜的海带,对其携带的菌群经过多次富集,得到相对稳定的具有海带降解效果的菌群。通过菌种的分离、纯化从而得到30株菌。对其进行16S rDNA测序及筛选,从而选择降解效果好的菌株及菌种组合。利用果胶或海藻酸钠为唯一碳源的选择性培养基挑选含有果胶酶、海藻酸裂解酶的菌株;通过测定海带发酵液中海藻酸含量及相应菌种的单菌及复合菌降解海带效果选出降解效果较好的单菌及复合菌。结果表明,菌种1-2及含有1-2和3-10的复合菌降解海带效果均在85%以上,其中1-2和3-10复合菌的降解效果为95%,效果较好。而复合菌的降解除几个效果差的外,其它基本达到70%以上,效果相对好于单菌的。(2)海带发酵液检测和成分分析海藻寡糖的粗提取:通过95%的乙醇溶液以粗提取海藻寡糖,并通过3,5-二硝基水杨酸比色法测定海带发酵液中还原糖和总糖含量以确定海藻寡糖的提取情况。由实验可知,1-2和3-10复合菌发酵20%海带的海带发酵液在提取前的还原糖和总糖含量分别为8.76、25.59g/ml,结果最好,经过海藻寡糖的粗提取,海带发酵液中都几乎不含还原糖和总糖,因此通过上述方法可提取出海藻寡糖。薄层层析:实验通过对展开剂和显色剂的分析可知,在展开剂正丁醇:乙酸乙酯:异丙醇:冰乙酸:水=2:6:6:4:1,显色剂使用20%硫酸-甲醇时条带效果比较好。海带发酵液的条带大致在蔗糖附近略靠下。由此可以推测出此海带发酵液中的海藻寡糖应大多为三糖。(3)海带发酵液对油菜种子萌发和生长的影响通过以稀释50、100、500、1000、2000倍海带稀释液,水为对照,处理油菜种子,观察油菜种子的发芽及发芽后根、茎的生长情况。实验表明,海带发酵液稀释1000倍处理的油菜种子发芽率最高为94.5%,比对照组高了58%;海带发酵液稀释2000倍处理的油菜芽根生长情况最好为4.44cm,比对照组高1.23cm;海带发酵液稀释2000倍处理油菜芽生长最好为5.44cm,比对照组高3.52cm。将育苗好的油菜种子种植于盆中,分别浇灌上水稀释倍数的海带稀释液,以水为对照,看油菜的生长状况。实验表明,海带发酵液稀释2000倍灌溉的油菜苗的茎长为7.90cm,比对照组高2.54cm,长于其他稀释液的种子。综上述结果可知,海带发酵液稀释2000倍的对油菜种子的促生作用最好,其次为1000倍的处理组。(4)海藻发酵液对土壤微生物菌群及植物生长的影响将育苗好的油菜种子种植于装有接种土壤菌悬液的无菌土的盆中,浇灌不同的海带稀释液,以水为对照,看油菜的生长状况及土壤微生物的生长状况。实验表明,油菜植株在1500稀释度生长比较好,植物植株大一些;土壤中放线菌、细菌和真菌均为海带发酵液稀释1500倍时的菌数最多分别为2.34×10~7,5.7×10~6,3.14×10~7个,说明此稀释度对土壤微生物的生长促进作用比较明显。
