作者毕可(2019)在《重组玉米赤霉烯酮降解酶的表达》一文中研究指出:玉米赤霉烯酮(Zearalenone)是一种主要由镰刀菌产生的具有假雌激素作用的霉菌毒素,会对动物的生殖系统造成损伤,影响生育功能。玉米是重要的粮食及饲料来源,同时也是受到玉米赤霉烯酮污染的主要农作物,近年来玉米赤霉烯酮的污染范围和程度有扩大上升趋势,因此如何降解受污染农作物中玉米赤霉烯酮正成为亟需解决的安全和经济问题。本实验采用的生物方法是酶法降解,选用一种玉米赤霉烯酮内酯水解酶,该酶能够打开玉米赤霉烯酮分子中的内酯环,使得开环后的玉米赤霉烯酮不再能与雌激素受体结合,从而起到毒素降解的作用。本实验成功在毕赤酵母中表达了一段来源于粗糙脉孢菌的玉米赤霉烯酮内酯水解酶基因,该基因全长888 bp,表达产物含有296个氨基酸,具有三联催化位点,属丝氨酸蛋白酶家族。通过电转化成功将该基因转入毕赤酵母中,获得重组毕赤酵母,并对该酶进行了摇瓶发酵和30L发酵罐发酵。甲醇诱导72 h后,重组毕赤酵母的摇瓶产量为40 U/ml;在摇瓶发酵之后还进行了30L发酵罐发酵,在发酵90 h,亦即甲醇诱导72 h时发酵罐酶活达到最大,为290.6 U/ml,验证了该酶进行大规模发酵生产的可行性。将该酶纯化后检测了其酶学性质,该酶最适pH 8.0,最适温度45℃,对玉米赤霉烯酮的Km值为38.63±5.868μM,Vmax为23.8μM/s/mg,对浓度为5 mM的Zn2+、Mg2+和Ca2+等离子具有较好的耐受性,对EDTA和SDS、Mn2+的耐受性较差。当处于pH 6.0-8.0,37℃的环境条件时,酶活可以稳定存在1 h,24 h后酶活仍保持近一半。该酶在对10μg玉米赤霉烯酮纯品进行降解时,反应10 min可降解82%的玉米赤霉烯酮,反应15min则可降解99%的玉米赤霉烯酮,液质联用检测产物为无毒物质。实验还研究了该酶在常见饲料原料和发酵麸皮中的玉米赤霉烯酮降解效果。其在DDGS、玉米副产物及玉米皮中降解实验中的降解率分别可以达到70.9%、88.9%和94.7%;其在发酵麸皮中的降解率最高可达51%,且加入发酵菌粉比发酵菌液更有利于降解。此外还尝试了对该酶应用固定化技术,提升了该酶对pH和温度的耐受性并且可以多次循环使用。其中以活化CNBr琼脂糖珠和活化NH2琼脂糖珠为载体的固定酶表现最佳,其在pH 5.0条件下相较于游离酶能够保持更多的酶活,分别为52.5%和56.2%;在60?C条件下的酶活分别为50.9%和32.4%。本实验对该酶进一步在实际生产中的应用有重要指导作用。
