作者栾天翔(2019)在《金属诱导Fmoc-氨基酸超分子水凝胶的研究》一文中研究指出:超分子自组装化学突破了传统的分子化学基于共价键的研究领域,通过精确调控非共价键作用力(氢键作用,主客体作用,π-π堆积作用等)制备有序聚集体,在近年来引起了广泛关注。本论文以单一及多元Fmoc-氨基酸为研究对象,研究了其在金属离子诱导下的超分子水凝胶的微观形貌,力学强度,并提出了可能的组装机理。同时探索了这类超分子水凝胶的潜在应用。论文主要包括三部分:第一章,简单介绍了超分子化学与超分子组装的基本概念与几种常见的超分子结构,以及基于芳香基氨基酸及多肽的超分子水凝胶体系的特性与功能应用。第二章,主要研究了金属离子诱导Fmoc-丙氨酸水凝胶的制备与功能。芳基多肽由于自身的π-π堆积作用和氢键作用,是一种良好的自组装基元分子。但芳基氨基酸由于氢键结合位点较少,难以通过自组装形成稳定的聚集体。我们发现,利用Cu2+,Eu3+等金属离子诱导Fmoc-丙氨酸可以形成性质稳定的水凝胶结构,通过扫描电镜与透射电镜等手段观测凝胶的微观形貌,发现Cu2+诱导水凝胶的微观形貌为0.5 μm左右的片状结构,Eu3+诱导水凝胶则组装为直径在50-300 nm的一维纳米纤维,当两种金属离子按等比例混合后诱导的水凝胶组装形貌转变为交联的3D网络,平均直径大约为20-100 nm。通过流变学测试表征了三种水凝胶的机械力学强度,发现三种凝胶都是力学强度较高的稳定水凝胶,屈服应力值都超过了 1000Pa。基于红外光谱,紫外光谱和小角X射线散射图谱的分析我们提出了水凝胶可能的组装机理,首先两个Fmoc-Ala形成反平行重叠π-π堆积的双层结构单元,之后在金属离子与羧酸根的配位作用及酰胺键之间的氢键作用下,继续形成层状结构,在三种超分子力的共同作用下,生成一维纳米纤维,进而以缠绕堆叠的方式组装成片状或纤维凝胶网络。令人意外的是,这种超分子水凝胶体系对于含有芳香基团和氨基的分子具有独特的溶蚀响应性,当把不同的氨基化合物或芳香化合物加入到凝胶中时,凝胶会发生特异性的溶蚀现象,且溶蚀的速率与溶蚀后的形貌均不相同。相信这种独特的客体响应水凝胶在分子识别和预封装材料中有着潜在的应用前景。第三章,主要研究了金属离子与复合Fmoc-氨基酸的共组装行为与组装体的形貌控制。我们首先研究了 Zn2+与单一 Fmoc-Ala或Fmoc-Val的超分子组装情况,Zn2+与Fmoc-Ala会组装为不稳定白色凝胶,凝胶会在短时间内坍塌生成浑浊溶液,在扫描电镜与透射电镜下可观察到数微米的片状聚集体。Zn2+与Fmoc-Val在一定的浓度范围内可组装成稳定的黄色凝胶,微观形貌为20 nm左右的纤维构成的交联网络。之后我们将Fmoc-Ala与Fmoc-Val按一定比例配置成混合溶液后再利用Zn2+诱导氨基酸形成超分子聚集体,发现当两种氨基酸的配比为1:1成功制得稳定的透明水凝胶,在扫描电镜下,可观测到透明水凝胶的微观形貌为纳米纤维与纳米球并存的结构。这说明复合Fmoc-氨基酸与Zn2+的超分子组装方式与单氨基酸体系不同。在流变学测试下我们发现,调控两种氨基酸的不同配比可以制得力学强度不同的稳定水凝胶体系。相信这种基于复合Fmoc-氨基酸的金属诱导水凝胶体系可以为我们理解多元超分子组装提供有力的支持。
