作者金丽颖(2019)在《抗黄瓜棒孢叶斑病放线菌的筛选及Streptomyces inhibens多相分类研究》一文中研究指出:黄瓜棒孢叶斑病是由多主棒孢(Corynespora cassiicola)侵染引起的,是近期威胁黄瓜生产的重要流行病害,造成黄瓜的大面积减产,寻找有效的防治措施是促进当前黄瓜种植产业可持续发展的主要任务。目前关于黄瓜棒孢叶斑病的防治方法主要还是采用化学防治,但由于该病原菌易变异,且长期频繁地施加化学试剂如杀菌剂等,又会导致该病原菌产生抗药性,更会引发食品安全和环境问题,因此黄瓜生产特别需要低毒、无害、环境友好型的防治方法。放线菌是一种分布广、种类多、功能性强的微生物,所以近年来常选用放线菌防治植物病害。本研究以黄瓜根际土壤及伴生小麦根际土壤为分离对象,采用多种培养基对放线菌进行分离纯化,通过抗黄瓜棒孢叶斑病原真菌及其他植物病原真菌和抗细菌活性筛选得到一株活性较好的链霉菌Streptomyces NEAU-J2,通过离体叶片和盆栽试验研究了该菌株对黄瓜棒孢叶斑病的防治效果并对其次级代谢产物进行分离,。同时,对得到的另一株活性放线菌新种进行多相分类学研究。本试验主要结果如下:(1)通过五种培养基(GS、HV、AAG、CPA、DPA)共分离出262株放线菌,其中119株分离自黄瓜根际土壤,其余143株分离自伴生小麦根际土壤。(2)通过抗黄瓜棒孢叶斑病病原菌活性初筛和复筛后,有32株放线菌对黄瓜棒孢叶斑病病原菌抑制率达到20%以上,其中有16株抑制率达到了40%以上;选取了抑菌活性最好的10株放线菌进行广谱的抑菌活性测定,发现菌株NEAU-J2具有较好的广谱抑菌作用。(3)黄瓜离体叶片和盆栽试验表明,用NEAU-J2孢子悬液及发酵滤液分别处理黄瓜叶片后,对黄瓜棒孢叶斑病有很好的防治效果。其中发酵滤液对黄瓜棒孢叶斑病的防效最好,在离体叶片和盆栽试验中的防效分别为81.2%和82.1%。(4)通过培养特性分析,菌株NEAU-J2培养特征与链霉菌属吻合,初步确定菌株NEAU-J2属于链霉菌属。此外功能特征分析表明,NEAU-J2发酵液对黄瓜棒孢叶斑病的抑制作用具有良好的紫外线稳定性,且菌株NEAU-J2可以产生几丁质酶,单位菌落透明圈直径为16.2mm。(5)对菌株NEAU-J2的次级代谢产物进行分离,初步得到三个已报道的化合物,分别为17-O-demethyl-geldanamycin、pteridic acid hydrate和pteridic acid F,其中pteridic acid hydrate对黄瓜棒孢叶斑病的抑制作用最好,当pteridic acid hydrate浓度为5 mg?mL-1时,对黄瓜棒孢叶斑病的抑制率为56.3%。(6)本试验通过对黄瓜棒孢叶斑病抑制率最好的10株放线菌测序发现了一株放线菌新种NEAU-D10,采用多相分类学方法分别从分子水平、形态学水平、化学分类水平、生理生化水平及全基因组分析对其进行研究。确定这株放线菌属于链霉菌属,并将其命名为Streptomyces inhibens。
