作者何晓蓓(2019)在《丁香提取物微乳的质量控制及稳定性研究》一文中研究指出:丁香(Caryophylli Flos)为桃金娘科植物丁香Eugenia caryophyllata Thunb.的干燥花蕾。具有温中降逆、补肾壮阳的功效。研究表明,丁香具有抗氧化、抗菌和杀螨等作用,含挥发油14%20%,其中丁香酚含量78%95%。本试验在前期研究基础上,对丁香药材、丁香提取物、丁香提取物微乳的质量控制及丁香提取物微乳的稳定性进行研究。取得以下结果:(1)丁香药材的质量控制A.鉴别:照《中国兽药典》2015版二部中丁香药材鉴别,取供试品溶液、对照药材溶液、丁香酚对照品溶液各5μL,点于同一硅胶G薄层板上,以石油醚(6090℃)-乙酸乙酯(9:1)为展开剂,展开,取出,晾干,喷以5%香草醛硫酸溶液,在105℃下加热至斑点显色清晰。供试丁香药材色谱中,在与丁香酚对照品和对照药材色谱相应的位置上,显相同颜色的斑点。B.含量测定:建立了丁香药材的高效液相色谱含量测定方法,其色谱条件为:色谱柱Welchrom C18(4.6×250nm,5μm),流动相:甲醇-水(55:45),流速1mL/min,柱温40℃,检测波长282nm,进样量20μL。丁香酚在0.019940.13958mg/mL浓度范围内与峰面积呈良好线性关系,线性方程为y=23045083.4647x+23918.2857,r=0.9998,平均加样回收率为101.46%,RSD=0.60%。测得供试丁香药材中丁香酚含量为15.52%。说明该含量测定方法专属性强,重复性好。(2)丁香提取物的质量控制A.鉴别:建立了丁香提取物的薄层色谱鉴别方法。具体是,照《中国兽药典》2015版二部中丁香药材的薄层色谱法鉴别,取供试品溶液10μL,对照药材溶液、丁香对照品溶液各5μL,点于同一硅胶G薄层板上,以石油醚(6090℃)-乙酸乙酯(5:1)为展开剂,展开,取出,晾干,喷以10%硫酸乙醇溶液,在105℃下加热至显色,在紫外灯(365nm)下检视,供试提取物色谱中,在与丁香酚对照品和对照药材色谱相应的位置上,显相同颜色的荧光斑点。该方法耐用性强。B.含量测定:取丁香提取物约0.06g,精密称定,加甲醇50mL,称定重量,超声提取20min,放置室温后,补足减失的重量,摇匀,滤过,取续滤液。照丁香药材质量控制研究中建立的高效液相色谱法测定。测得供试丁香提取物中丁香酚含量为5.87%。(3)丁香提取物微乳的质量控制A.鉴别:称取丁香提取物微乳20.0g,加无水硫酸钠2.0g,超声破乳30min,加石油醚(3060℃)20mL萃取3次,合并,室温挥干,加乙醚2mL溶解即得丁香提取物微乳溶液,同法制得空白微乳溶液。取微乳、空白微乳各20μL,照对丁香提取物质量控制研究中建立的薄层色谱法鉴别,丁香提取物微乳色谱中,在与对照药材和丁香酚对照品色谱相应的位置上,显相同颜色的荧光斑点,阴性对照无干扰。B.含量测定:取丁香提取物微乳约3g,精密称定,加甲醇定容至50mL,照丁香药材质量控制中建立的高效液相色谱法测定,空白微乳无干扰。测得供试丁香提取微乳中丁香酚含量为0.05%。(4)丁香提取物微乳的稳定性强光照射试验、高温试验、加速试验和长期稳定性试验结果表明,丁香提取物微乳,外观呈黄褐色,均一透明,无分层现象,且丁香酚含量未见明显变化。说明该制剂在室温避光条件下,保存6个月稳定。

ding xiang (Caryophylli Flos)wei tao jin niang ke zhi wu ding xiang Eugenia caryophyllata Thunb.de gan zao hua lei 。ju you wen zhong jiang ni 、bu shen zhuang yang de gong xiao 。yan jiu biao ming ,ding xiang ju you kang yang hua 、kang jun he sha man deng zuo yong ,han hui fa you 14%20%,ji zhong ding xiang fen han liang 78%95%。ben shi yan zai qian ji yan jiu ji chu shang ,dui ding xiang yao cai 、ding xiang di qu wu 、ding xiang di qu wu wei ru de zhi liang kong zhi ji ding xiang di qu wu wei ru de wen ding xing jin hang yan jiu 。qu de yi xia jie guo :(1)ding xiang yao cai de zhi liang kong zhi A.jian bie :zhao 《zhong guo shou yao dian 》2015ban er bu zhong ding xiang yao cai jian bie ,qu gong shi pin rong ye 、dui zhao yao cai rong ye 、ding xiang fen dui zhao pin rong ye ge 5μL,dian yu tong yi gui jiao Gbao ceng ban shang ,yi dan you mi (6090℃)-yi suan yi zhi (9:1)wei zhan kai ji ,zhan kai ,qu chu ,liang gan ,pen yi 5%xiang cao quan liu suan rong ye ,zai 105℃xia jia re zhi ban dian xian se qing xi 。gong shi ding xiang yao cai se pu zhong ,zai yu ding xiang fen dui zhao pin he dui zhao yao cai se pu xiang ying de wei zhi shang ,xian xiang tong yan se de ban dian 。B.han liang ce ding :jian li le ding xiang yao cai de gao xiao ye xiang se pu han liang ce ding fang fa ,ji se pu tiao jian wei :se pu zhu Welchrom C18(4.6×250nm,5μm),liu dong xiang :jia chun -shui (55:45),liu su 1mL/min,zhu wen 40℃,jian ce bo chang 282nm,jin yang liang 20μL。ding xiang fen zai 0.019940.13958mg/mLnong du fan wei nei yu feng mian ji cheng liang hao xian xing guan ji ,xian xing fang cheng wei y=23045083.4647x+23918.2857,r=0.9998,ping jun jia yang hui shou lv wei 101.46%,RSD=0.60%。ce de gong shi ding xiang yao cai zhong ding xiang fen han liang wei 15.52%。shui ming gai han liang ce ding fang fa zhuan shu xing jiang ,chong fu xing hao 。(2)ding xiang di qu wu de zhi liang kong zhi A.jian bie :jian li le ding xiang di qu wu de bao ceng se pu jian bie fang fa 。ju ti shi ,zhao 《zhong guo shou yao dian 》2015ban er bu zhong ding xiang yao cai de bao ceng se pu fa jian bie ,qu gong shi pin rong ye 10μL,dui zhao yao cai rong ye 、ding xiang dui zhao pin rong ye ge 5μL,dian yu tong yi gui jiao Gbao ceng ban shang ,yi dan you mi (6090℃)-yi suan yi zhi (5:1)wei zhan kai ji ,zhan kai ,qu chu ,liang gan ,pen yi 10%liu suan yi chun rong ye ,zai 105℃xia jia re zhi xian se ,zai zi wai deng (365nm)xia jian shi ,gong shi di qu wu se pu zhong ,zai yu ding xiang fen dui zhao pin he dui zhao yao cai se pu xiang ying de wei zhi shang ,xian xiang tong yan se de ying guang ban dian 。gai fang fa nai yong xing jiang 。B.han liang ce ding :qu ding xiang di qu wu yao 0.06g,jing mi chen ding ,jia jia chun 50mL,chen ding chong liang ,chao sheng di qu 20min,fang zhi shi wen hou ,bu zu jian shi de chong liang ,yao yun ,lv guo ,qu xu lv ye 。zhao ding xiang yao cai zhi liang kong zhi yan jiu zhong jian li de gao xiao ye xiang se pu fa ce ding 。ce de gong shi ding xiang di qu wu zhong ding xiang fen han liang wei 5.87%。(3)ding xiang di qu wu wei ru de zhi liang kong zhi A.jian bie :chen qu ding xiang di qu wu wei ru 20.0g,jia mo shui liu suan na 2.0g,chao sheng po ru 30min,jia dan you mi (3060℃)20mLcui qu 3ci ,ge bing ,shi wen hui gan ,jia yi mi 2mLrong jie ji de ding xiang di qu wu wei ru rong ye ,tong fa zhi de kong bai wei ru rong ye 。qu wei ru 、kong bai wei ru ge 20μL,zhao dui ding xiang di qu wu zhi liang kong zhi yan jiu zhong jian li de bao ceng se pu fa jian bie ,ding xiang di qu wu wei ru se pu zhong ,zai yu dui zhao yao cai he ding xiang fen dui zhao pin se pu xiang ying de wei zhi shang ,xian xiang tong yan se de ying guang ban dian ,yin xing dui zhao mo gan rao 。B.han liang ce ding :qu ding xiang di qu wu wei ru yao 3g,jing mi chen ding ,jia jia chun ding rong zhi 50mL,zhao ding xiang yao cai zhi liang kong zhi zhong jian li de gao xiao ye xiang se pu fa ce ding ,kong bai wei ru mo gan rao 。ce de gong shi ding xiang di qu wei ru zhong ding xiang fen han liang wei 0.05%。(4)ding xiang di qu wu wei ru de wen ding xing jiang guang zhao she shi yan 、gao wen shi yan 、jia su shi yan he chang ji wen ding xing shi yan jie guo biao ming ,ding xiang di qu wu wei ru ,wai guan cheng huang he se ,jun yi tou ming ,mo fen ceng xian xiang ,ju ding xiang fen han liang wei jian ming xian bian hua 。shui ming gai zhi ji zai shi wen bi guang tiao jian xia ,bao cun 6ge yue wen ding 。
