作者杨瑜(2019)在《宽频电磁脉冲对人羊膜上皮细胞凋亡及胞内钙离子的影响》一文中研究指出:前期研究发现,宽频电磁脉冲(Wide-band electromagnetic pulse,WEMP)会对大鼠的心脏、肝脏、睾丸等组织造成细胞凋亡损伤。为了进一步探索WEMP辐照参数对于细胞凋亡的损伤规律和凋亡机制,本文以人羊膜上皮细胞(amniotic epithelial cells,AECs,或FL细胞)为实验对象,研究了WEMP的两个主要的辐照参数(场强和脉冲个数)与离体细胞凋亡之间的关系以及与凋亡相关的细胞钙内流作用关系,拟为WEMP导致细胞损伤的生物物理模型的建立奠定前期基础。本实验首先利用XFdtd 7.3.2对细胞辐照体系模型在WEMP辐照情况下的电磁暴露剂量理论计算;采用流式细胞术检测Annexin V-FITC/PI染色确定受照后细胞凋亡情况;采用Fluo-4 AM等荧光染色检测了受照后细胞内钙离子分布及浓度变化;采用DPH荧光偏振法检测了细胞膜流动性变化;采用蛋白质印迹法(western blotting)检测了细胞中相关蛋白的含量变化;利用SPSS 23.0软件对实验数据进行了统计分析,获得了WEMP不同参数与细胞凋亡之间的关系。实验结果表明:1.在实验所采用的WEMP辐照条件下,作用于培养皿中的场强约为外场强的7%,经计算可得本实验条件下细胞层的最大温升约为7.04×10-3℃,以非热效应为主。2.WEMP辐照后,AECs的凋亡与场强(E)之间为Logistic函数关系,与脉冲个数(N)呈指数关系,N与E之间存在交互作用,E为引起细胞凋亡的主因。3.WEMP辐照能够改变细胞内钙离子浓度,当细胞外钙浓度较高时,WEMP能够使胞浆中钙浓度上升,且场强越大,脉冲个数越多,造成短时间内胞内钙离子浓度上升越明显,与细胞凋亡之间存在显著的相关性;当细胞外钙低于正常浓度时,WEMP能够使胞浆中的钙浓度下降,且场强越大,脉冲个数越多,造成短时间内胞内钙离子浓度下降越明显。4.WEMP辐照对AECs膜流动性无显著影响,胞浆内钙离子浓度上升可能由于WEMP影响了细胞膜上离子通道的开放。5.WEMP辐照引起了胞浆内钙离子的升高,钙离子在复杂的细胞凋亡中扮演重要角色,本实验中caspase 9被剪切活化提示钙离子有可能参与激活了细胞凋亡的内源性通路。
