作者陈永卓(2019)在《噪声对黑线毛足鼠焦虑行为的影响及其分子机制》一文中研究指出:高速公路噪声作为一种环境刺激,可以激活HPA轴,CRHR1受体在HPA轴对外界刺激产生应答过程中起关键作用。我们探究了急慢性噪声对黑线毛足鼠行为及下丘脑中CRHR1表达的影响。将40只成年黑线毛足鼠随机分为以下8组,每组5只:(1)雌性急性对照(F AC,Female Acute Control)、(2)雌性急性85dBA处理(F A85,Female Acute 85dBA exposure);(3)雄性急性对照(M AC,Male Acute Control,)、(4)雄性急性85dBA处理(M A85,Male Acute 85dBA exposure);(5)雌性慢性对照(F CC,Female Chronic Control)(6)雌性慢性85dBA处理(F C85,Female Chronic 85dBA exposure);(7)雄性慢性对照(M CC,Male Chronic Control)、(8)雄性慢性85dBA处理(M C85,Male Chronic 85dBA exposure)。急性处理组使用85dBA高速公路噪声处理4h,慢性处理组在相同噪声条件下每天4h,持续30天。下丘脑中CRHR1表达量使用实时荧光定量PCR检测。通过红外旷场实验、明暗箱实验以及高架十字迷宫实验检测噪声对黑线毛足鼠行为的影响,急性噪声处理后行为学实验仅进行一次,慢性噪声处理后行为学实验每5天进行一次。急性噪声使雌雄黑线毛足鼠产生轻微的焦虑行为,对下丘脑CRHR1的表达无显著影响,慢性噪声使雄性个体表现出一定程度异常冲动和亢奋的低焦虑行为并使下丘脑中CRHR1表达显著降低,但慢性噪声对雌性行为的影响要小于雄性个体且对雌性下丘脑中CRHR1表达没有显著影响。相关性分析表明,下丘脑CRHR1表达量与中央区域停留时间呈极显著负相关r=-0.579,P<0.01,与中央区域运动距离呈显著负相关r=-0.466,P=0.014,但是与其他行为学指标的相关性不显著。本研究表明,急慢性噪声均可以导致黑线毛足鼠行为变化,并且慢性噪声可以导致雄性个体下丘脑CRHR1表达降低,本研究为掌握CRHR1在噪声调控动物行为中的作用效应提供理论基础。
